In this paper , the molecular structure , characteristic of molecular spectra , methods for preparation and the applicable areas of metal phthalocynine and metal free phthalocyanine were commented . The developments of the probes of protein and some major methods used in the analysis of protein in recent years were reviewed.Tetra sulfonic phthalocyanine zinc (ZnPcS4) was synthesized by using 4-sufo-phthalic acid as fragment, (NH4)2MoO4 as catalyst. Then it was used as a kind of protein probe, the interaction of ZnPcS4 with BSA was studied by UV/Vis and fluorescence spectroscopy method. The result indicated that the interaction of ZnPcS4 with BSA was in accord with model of phase distribution, the combination of BSA with Z11PCS4 was considered as static electricity forces, the binding constant of ZnPcS4 with BSA was K=7.82x10~5. Fluorescence spectroscopy showed that the binding constant K=2.02X10~5 which was basically accorded with the binding constant determined by UV/Vis .A series of tetra-carboxylic metal phthalocyanine(zinc , aluminum , manganese , nickel , copper , cobalt) and octa-carboxylic metal phthalocyanine(zinc , aluminum , manganese , cobalt )were synthesized. Then they were used as a kind of protein probe. The binding constant of carboxylic metal phthalocyanine with BSA was determined by fluorescence spectroscopy method. The results indicated that there was no much differences among the different metal ions and number of carboxylic substituents for the binding constant of carboxylic phthalocyanine with BSA. The binding site ofcarboxylic phthalocyanine with BSA was studied by competitive binding method, the result showed that the carboxylic phthalocyanine only binded with BSA at site I.
