Aging and longevity is the most difficult complex subject of human science research,and aging is the result of gene and environment two aspects joint action. Among them, the gene is the key factors internal decided to the ageing of the cells, tissues and organs of old age, and the old and the overall aging system. How to control the activity of aging related genes from the source of state, aging initiative, slow or on intervention consenescence of the initiation and development of the process, is the key to the research.This research collection and analysis the current proven C.elegance longevity genes to build into C.elegance longevity genes local network. From the perspective of gene network can reflect the inherent law of life itself better than ever before on the single isolated gene. C.elegance be choosen model organisms, and two feeding schemes that are different kinds of lactic acid bacteria and branched-Chain Amino Acids and their combination. Established by using different nutrients feed C.elegance mode, and screening the best combination of longevity.Using real-time PCR detection expression changes of gene that C.elegance of using the optimal combination in the growth of the several stages , the genes that we are choosen is the local longevity genes network of C.elegance.Of course, the current for complex biological properties research there is no fixed research strategies and methods, many environmental factors and sensitive gene interactions can be influenced on the biological properties, this process is also a dynamic circulation.The test results show that the part of the lactic acid bacteria in the NGM flat can't grow very well, lead to the live and transfer of C.elegance is difficulty. So the test directly to these strains. Final strains of feeding C.elegance is 11 plants, respectively, No.1,No.8, No.5, No.1936, No.119-10, No.6-3-1,No.7-3-1,No.6-3-2,No.7-3-2, No.6-4, No.7-4. In the 11 strains of lactic acid bacteria of we test,not all of the lactic acid bacteria on life have effect, with only four strains have an effect on the life of C.elegance and that are No.1936,No.6-3-2,No.6-4,No.7-4 and all of the lactic acid bacteria not influence in the largest C.elegance life , but reflected in the reduce mortality in the earlier stage.The second nutrients is branched-Chain Amino Acids, nutrients directly add to medium, still use E.coli OP50 to feed C.elegance. Total choose three kinds of amino acids, which are valine, leucine and isoleucine, each amino acid concentration, with three of concentration are 0.1 times,1 times and 10 times. Then choose the optimal combination from amino acid . The results show that a combination of amino acids no better than a single amino acids on C.elegance life influence. In all the nutrients test,0.1 times valine is the best.And the effect is not only extension the biggest life of C.elegance, but also effect on the reduction in death in the whole life of C.elegance.Through the real-time PCR analysis 14 the gene expression around the metabolism of valine,and the results show that the expression of SPTL-2, cyp-33 E1, C16A3. 10, odc-1, C2.8 H8.11 and alh-13 are increased. Expression of Unc-25 is decreased. C05D2.3 has a different effect at different stage,the expression is reduction in the early and raise in the middle. From the differences in the different genes, we can further inferences of longevity genes related function and the related level.Also can speculate that did not report the genes could be longevity genes.By constructing the longevity gene expression database,can support longevity data detection of C.elegance.
