The major object of this paper is to isolate and culture mouse embryonic germ (EG) cells derived from primordial germ cells (PGCs) as well as to identify the EG cells from various aspects. Some factors influencing the efficiency of the isolation and culture of mouse EG cells have been discussed. Our work may be useful to further study the biology of mouse EG cell and establish the EG cell lines. The results obtained were as follows: 1. Isolation and culture of mouse primordial germ cells was studied in 8.5~12.5dpc fetuses. One EG cell line was maintained the undifferentiated state for 9 passages. 2. Experiment was performed to compare the effect of different methods on isolation of mouse EG cells. The result suggested that the dispersal effect of "several times digest"was better than "one time digest". 3. Comparing the effect of different dispersal methods on the subculture of mouse EG cells, the method of "digest-mechanical dispersal", "digest-continuous dispersal"and "digest-blowing dispersal"were fine to be used in the propagation of EG cells, but the"digest-continuous dispersal"and the "digest-blowing dispersal"is more efficient for the work of EG cells passages. 4. When mouse primordial germ cells were cultured in three different cell medium, all can clone primary EG cells. Mouse fetus liver cells condition medium, which can maintain the survivance and proliferation of mouse EG cells, is better than the medium contained no cytokine, but worse than EG cell medium with 1000IU/mL LIF. 5. When mouse primordial germ cells were cultured in the cell medium containing 17βestradiol is better than the control group, having more EG cell colonies with undifferentiated phenotype. In subculture, the colonies from the 17βestradiol group is cultured more passages. 6. We found that the mouse fetuses with different sex both can be used to isolate the primordial germ cells, but the efficiency is different, the male is better than female fetuses for primary and subculture of PGCs or EG cells.
