With the progress of urbanization and living standard, the urban life sewage discharge is becoming more and more serious, so the city sewage treatment is much more important. Now, the main way of city sewage treatment uses microbe in active sludge to purify water. These microbes can not only purify the water, but also predict Water Quality; What's more, they can help to make a scientific adjustment about process control. The protozoan and metazoan play an important role in microbe. Therefore, identifying protozoan and metazoan in micro photos is very necessary.This paper, as a part of project called "image process and recognition of common protozoa and metazoan in activated sludge system", put the focus on the count of the protozoa and metazoan. The main work and achievements are as follows:Due to the color difference between protozoa and background in micrograph, the paper explores two different image preprocessing algorithm to improve image quality and lay the foundation for subsequent counting. One is designed for the most of the images, in other words, most images can achieve desired results in this way; The other algorithm is designed for special cases, which the color between the protozoa and background is almost the same, and it can't be extracted by the first way. It combines human interaction and Hough transform to solve the problem. Then, the paper studies the traditional counting methods and proposes a new way to make counting result fixed for overlapped microbes, which based on both area and circularity.Above all, the paper gives the implementations about the protozoa and metazoan's counting, what's more, it makes the foundation for further research.
