Pinus koraiensis belongs to sonko loose genus, it is expensive and rare , one of the second-class national key protection plants. It's mainly distributed in the northeast ,Changbai mountain area and Xiaoxinganling area. It's one of the oldest gymnosperms tree, emerged about 25-23 million years ago. During its evolution process, Pinus koraiensis experienced the whole quaternary climate variation.This paper is based on the distribution of the three northeast provinces in our country , 25 natural Pinus koraiensis populations,and 125 individuals are for material. Using the mitochondrial DNA primer on the PCR nad5 by PCR products, the method for determining direct sequencing the mitochondrial DNA (mtDNA) nad5 the coding region series, and the molecular biology software on each sequence to compare, record mutations, summary sheet times in order to construct Phylogenetic tree according to Haplotype. On the analysis of natural Pinus koraiensis system development in northeast natural geographical mode will be a Haplotype to type system with all kinds of the evolutionary tree of geographical position combined.Using the software Primer5.0 to design a pair of amplification primer nad5,125 natural Korean pine individuals to enlarge and direct sequencing of PCR products. Sequencing DNA sequence and get 122 bar in accordance with these sequences FASTA format is arranged, Clustal X Ver1.81 software to use its differences, delete its credibility after the footage nad5 determined by primer amplification than a and can be used in the fragment length in 436bp - 437bp,did not show obvious length polymorphism. T, C, A, G four nucleotide for the average ratio of 24.8 %,25.9%, 26.4%, 22.9%, G, C content was 48.8% and A,T content was 51.2%, G, C content was slightly lower than A,T content, much of non-coding areas have this feature.By analyzing and comparing 122 sequencings are got, variation sites were identified 10, defined 12 mtDNA Haplotype, it's named HA-HL. HA is called Ancestors of Haplotype because Haplotype HA exists in all populations, it may be the most primitive times the haploid type. Part of Haplotype exists in multiple populations,so Haplotype is called sharing model, this phenomenon proves different populations shared genetic exchange. The rest of Haplotype for part of peculiar endemic species, called Haplotype. Using DnaSP4.10.9 software analyzes various provinces in group and Haplotype, it helps determine type diversity of Pinus koraiensis population to form natural glacial shelters. Finally using molecular evolution MEGA4.0 genetic analysis software with the floatation method and MP method, construct the natural Pinus koraiensis tree. A combination of these analysis in the process of the quaternary glacial period determined to temporary, China's natural shelter for Pinus koraiensis forest in Ji lin area of DST - the forest from QL (Dun hua forest of Hei longjiang province) and LSH - XX (Hai Lin category of forestry administration emerging forest, and presumably) of two populations. And presumably a refund period after the glacial but the spread of natural Pinus koraiensis forest proliferation path.The distribution of Pinus koraiensis is influenced by climate , this paper discusses the statistical studies on vegetation distribution change which lead by present climate, making reasonable strategies , but also suggesting the restoration and update for natural Pinus koraiensis feasibly.
