Drosophila is used extensively in the research of biomedicine and pharmacy. In medical studies, they are sually treated with reagents of interest and the changes in their organs, particularly their compound eyes, are then collected and analyzed. At present, these series of examine processes are still done manually,which makes the research very laborious and inefficient. This paper studies on the theories and technologies of auto-focusing in drosophila's compound eyes lesion discrimination.It has made some effective improvements in the following aspects:microscope auto-focusing image collection,drosophila's compound eyes lesion determination, drosophila's compound eyes lesion recognition. Based on the LabVIEW software,the microscope's auto-focusing is achieved. Image definition criterion function is the key for auto-focusing, in order to evaluate the practicality for auto-focusing,four methods were compared and analyzed.The parameters of sensitivity,consistency and responsiveness were tested and validated through experiments.The experiments and analysis show that the variance of gray-scale has not only focusing scale,but also good sensitivity and high responsivenes. This system applied variance of gray-scale as image definition criterion function.This paper optimized mountain-climb searching algorithm at auto-focusing algorithm. The optimized mountain-climb searching algorithm is more accurate and efficient compared with ordinary mountain-climb searching algorithm. The experrimental results show that it can realize fast and accurately automatic focusing.It settles the base for drosophila's compound eyes lesion tests. Based on image processing software Vision Assistant, it realizes three kind of extraction the contour line of the drosophila's compound eyes.The designed system has realized the quick analysis, the computation and provided results of the drosophila's compound eyes bad area position and the bad spot size. Finally, taken LabVIEW 8.6 software as the platform, the script that processing the single picture in Vision Assistant as the foundation, human-computer interface has been established, the automatic discrimination system of the drosophila's compound eyes lesion has been completed. These will be a new effective method to test the drug reaction of the subject (such as drosophila) participated the experiment.
