The image segmentation is to partition a picture into several segments according to its characteristics and the rules. The fit algorithm is the key of image segmentation and a little bit difficult. Colony image is very complex, so colony image processing is difficult.In this paper, a new segmentation method will be presented for the complex, unequal size, overlapping colony images. The study about the colony image processing includes the following parts:1. Image preprocessing. Image preprocessing technologies include image enhancement, filtering, edge detection. Pretreatment of the original colony image is to remove the noise in the image, smooth image and enhance the difference between target and background. It is foundation of image segmentation.2. Automatic multi-threshold segmentation. Based on the traditional threshold segmentation method, it segment the target image and background image again to segment more deeply.3. Gray contour extraction. Gray contour is used to the colony image segmentation. The Sobel operator is used to extracted gray contours form Multi-threshold segmentation. Finally, the gray contour lines are added to the colony image.4. Acceleration determination. According to the concept of acceleration in physics, it is determine the acceleration of contour lines near the pseudo-contour line to find the boundary. At the same time it is try to remove pseudo-boundary lines, and get the segmentation results.5. Colony image processing. Gray contour and acceleration determination are used in Colony image processing. The simulation results of Colony image processing are compared with the traditional methods results.
