Objective: To investigate the effect on thyroid axis hormone in nonalcoholic fatty liver disease(NAFLD) rats and the possible mechanism of aerobic exercise. Methods: 50 healthy male SD rats of 8 weeks, a random sample of 30 only by high-fat diet for 8 weeks to model nonalcoholic fatty liver in rats. The remaining 20 were selected as the normal control group. At 8 weeks, each group of 10 rats were randomly selected as the control group for 8 weeks(C8) and 8 weeks of high-fat diet group(M8), non-alcoholic fatty liver rat model was established by contrast detection. Then the normal rats and the successful model rats were divided into three groups: The 16 weeks normal control group(C16), 16 week high-fat diet group( M16) and high-fat exercise group(EM).The EM group were trained 60 min every day, without load swimming, training 6 days a week, a total of 8 weeks of training. Exercise for 8 weeks, using the ELISA method for the determination of TRH, TSH, T3, T4, rT3 and TBG.Results: 1) Optical microscope examination suggested that liver cells had no obvious fatty denaturation in C8, but 8 of 10 were found fatty degeneration of liver cells in M8, showing arranged in disorder, obvious vacuolization, ballooning degeneration. 2 of 10 were found a small amount of fatty degeneration in liver cells in C16.All the rat liver cell fatty degeneration is obviously visible in M16, showing hepatic sinus expansion and blisters in portal area, liver lobular inflammation. 6 of 9 were found fatty degeneration in liver cells in EM but no inflammation or necrosis, fibrous tissue hyperplasia. 2) TRH: there was no significant difference among the three groups.3) TSH level in M16 were slightly higher than that of C16 and EM group, but the three groups showed no significant difference.4) T3 level in M16 was significantly lower than that of C16. T3 level in EM was lower than that of C16 but no significant difference. T3 level in EM was higher than that of M16 but no significant difference. 5) T4 level in M16 was significantly lower than that of C16. T4 level in EM was higher than that of M16 but no significant difference. T4 level in EM was lower than that of C16 but no significant difference. 6) rT3 in M16 was significantly higher than that in C16. rT3 in EM was significantly lower than that of C16 and M16.Conclusions: 1) NAFLD model by 8 weeks of high-fat diet is successful.2) High fat- diet for 16 weeks reduce the thyroid function of NAFLD rats, showing that significantly higher of rT3, significantly reduced of T3 and T4.3) 8 weeks aerobic exercise can significantly reduce the degree of fatty liver of NAFLD rats,improve the levels of T3 and T4.
