Objective:Based on the period of the6weeks of different load rats swimmingtraining to build the sports models that the best swimming load cognitive and sportability through the Morris water maze and the flume experiment about behavior test,gene and protein level tested and checked the abroad research about the relationshipbetween PDGF-BB on the cognitive ability and sport ability, so that it can support theexperiment later on about the rats swimming training.Methods:50rats purchased from Kunming Medical University, and they weredivided into control group and excerise groups, and excerise groups can be dividedinto zero load group(S0group),1%load group(S1group),3%load group(S3group),and5%load group(S5group) by the load percent, each group10rats. Weighted therats every Monday early in the morning and recorded it. All rats for a week of fitnesstraining, then began to six-week experiment,1h/groups/day,5days/per week, OnSaturday test sports ability score,and rest on Sunday.Swimming tank do it by ourself:1.5*0.3*0.3(m), the water temperature is24±2℃.It began water maze experiment after six weeks training period, it’s include5days, the first four days for navigation experiment, test rats learning ability; The lastday for space exploration experiment, tested rats memory ability. The day after theBehavior test,we collected samples. The samples included hippocampus, cortex motor.Immunoblotting experiment and PCR experiment followed it.PCR experiment testrats’ gene expression level, Immunoblotting experiment tested rats’ proteinexpression level.Results and conclusions:1. There were no obvious difference among and within groups in the initial weightand adapt week.On the formal experiments during the six weeks, compared withcontrol group,the exercise groups’ weight was reduced, and had difference gradually.During the fifth week and the six week, compared with the S0group, the weight ofthe S3group is reduced significant. Compared the other exercise groups’, there had not statistical difference with S3group, although the S3group’s weight is minimum.2. Swimming training on cognitive function and regulated hippocampalPDGF-BB-related signal pathways downstream molecules.2.1In the navigation experiments, the average escape latency of rats with differentload exercise groups were declined with the increase of the number of training, andthe latency to a certain extent reached a plateau, basically did not change. In the fourdays experiments, on the first day and second day, the S1group’s escape latency wasthe shortest, while, on the third day and the fourth day, the S5group’s escape latencywas the shortest. The control group had always been the longest.There were noobvious difference among different load exercise group.2.2In the navigation experiments, from the rats fell into the water to find platform’sswim length to a certain extent to reflected the rat’s learning ability, it reflects the ratsto find a platform strategy. In addition to the second day of S3and S5group, othergroups’s swimming length gradually reduced with the increase of the number ofdays,it showed that the cognitive ability of rats was getting better and better. Therewere statistically significant along with the increase of the number of days betweenthe control group and the exercise groups.2.3In navigation experiments, with the increase of the number of test days, thenumber of rats found the platform in60s increased gradually, by contrary, theplatform was the number of rats not find the platform gradually reduce.Comparedwith the control group,the rats in exercise groups can easily find the platform.2.4In space exploration experiments, S5group in the target quadrant time percentageand distance percentage accounted for the largest share, it suggested that the S5grouprats has the best memory ability.2.5In the PCR experiments, PDGF-BB-related to cognition downstream signalpathways in rats is the RAS pathway and SRC pathway.However,there was notobvious difference among groups on the RAS pathway of PDGF BB, RAF1, MEK,ERK1’s mRNA level. On the SRC pathway, compared with the group S1andS3,SRC mRNA levels was declined in S5group.2.6According to the results of immunoblotting experiment,in the non-phosphorylationof ERK, compared with S3group, S0group’s protein expression was rised; compared with the group S1and S3,the S5group’s protein expression was rised; In thephosphorylation of ERK, compared with the group S0and S5,the C group’s proteinexpression was declined.3Swim training on athletic ability and regulated motor cortex PDGF-BB-relatedsignal pathways downstream molecules.3.1In the the flume experiment conducted on every Saturday,compared with thecontrol group,the exercise group’s swim length increased.On the fifth weekend,compared with the S0group,the S3group and the S5Group’s swim lengthincreased;On the sixth weekend, compared with the S0group,S1group’s swimlength increased.3.2In the navigation test, compared to every group’s swim speed, the first day’sswimm speed, compared with the S5group, the speed of the S1group is increasedsignificantly. On the sencond day’s experiment, compared with the S5group, the restgroups swim speed increased significantly.With the increase of the training day, thecontrol group and the exercise group can quickly learn to find the best way toplatform, although there were differences among groups,however, not statisticallysignificant.3.3The PCR experiments showed that:(1) PDGF-BB: compared with the S5group,the group C, S1, S5were fell, the difference was statistically significant;(2)PKB: Compared with the group S3and S5, the C group was rised;(3) PKC:comparedwith the S3group, the group S0, S1, S5were fell;(4) RAF-1: compared with S0, S5group was declined.(5) MEK-1:Compared with the C group, S1group was declined;Compared with S5group,the group C, S0and S3were rised;(6) ERK1: comparedwith S3group,the S5group was declined.(7) PI3K: Compared with the S0group,thegroupS1and S5were fell.(8) STAT3: Compared with the C group,the group S3andS5were fell.3.4Immunoblotting experiment, in the non-phosphorylation of ERK, comparedwith the C group, the groupS0and S5’s protein expression were rised; In thephosphorylation of ERK,compared with the group S3and S5, the group C and S0’s protein expression weredecreased. In the non-phosphorylation of STAT3, compared with group S1、S3and S5, the groupC’s protein expression was decline; Compared with the group S1and S3, the groupS0’s protein expression was rised. In the phosphorylation of STAT3, compared withgroup C, S0, S3and S5, of S5group. the group S1’s protein expression was rised.
