Purpose:To morphology and molecular biology techniques, observation of irritable bowel syndrome (IBS) rat model of fur, breathing, airway responsiveness (PC20 values), pathological changes in lung tissue, lung tissue substance P (Substence P , SP) content changes, protein expression, gene expression and protein expression of NK1 receptors and SP content in liver tissue, and explore the " the interior and exterior relationship between the lung and the largeintestine " the scientific meaning, and provide experimental evidence for the theory.Materials and methods:The 24 SPF Wistar male rats were randomly divided into control group and the IBS model group (the model group) two groups of 12. Cycle by 18 days to accept six different chronic stress stimulation (cold water swim, tail clip, ban water, fasting, electric shock, day and night reversed), while daily use of tape beam shoulder, forelimb and chest to restrict its scratch head and face of the binding method in rats with irritable bowel syndrome (D-IBS) pathological model. SPSS10.0 software using the experimental data t test analysis.Results:1 General Status: control rats, lively, shiny fur, breathing evenly, respiratory rate, an average of 74.21 times / min, etc.; model rats appeared diarrhea, irritability, aggressiveness, messy fur dull, breathing smooth, respiratory rate average stood at 109.64 times / min and significantly speed up the control group (P <0.01).2 light microscope: the control group alveolar structure is clear, continuous, alveolar wall integrity, and no infiltration of inflammatory cells; mod el group alveolar structure is not continuous, incomplete alveolar wall, alveolar expansion, occasional interstitial hemorrhage, inflammatory cell infiltration.3 PC20 values: model group, Paw airway pressure 20% higher concentration of histamine (PC20) was (0.06±0.02) and the control group (0.46±0.12) compared with decreased (P <0.01).4 SP content in lung tissue: model lung tissue SP content (298.62±12.58) and the control group (73.04±9.45) compared with significantly higher (P <0.01).5 SP content of liver tissue: model group, liver tissue SP content (51.27±8.51) and the control group (48.93±5.40) compared with no difference (P> 0.05).6 SP protein expression in lung tissue: model protein in lung tissue SP-positive index (PI value) (28.34±10.38) and the control group (16.58±6.43) compared with significantly higher (P <0.01).7 SP gene expression in lung tissue: model group, lung tissue SPmRNA /β-actin (gray) was 0.52±0.16 and control group (0.46±0.09) compared to upregulated (P <0.01).8 NK1 protein expression in lung tissue: model group were positive for NK1 protein index (PI value) (16.13±7.58) and the control group (10.56±4.23) compared with significantly higher (P <0.01).Conclusion:1 IBS mess dull fur rats, respiratory frequency is not smooth and speed up.2 IBS model rats significantly increased airway sensitivity.3 IBS rat model of lung tissue SP content increased, while liver tissue SP content were not significant.4 IBS rat model of lung tissue SP and the NK1 protein expression upregulated.5 IBS model of rat lung tissue SPmRNA upregulated.
