Objective:To prove the curative effect of electropuncture (acupuncted shenshu, geshu, baihuixue) and further for the functional mechanism on treating VD mice through the animal experiment.Methods:Part1. the effect of electropuncture on Praxiology of cerebral ischemia reperfusion injury miceThe healthy♂Kunming mice were randomly divided into four groups, the false surgical operation group, the model group, the xidezhen group, the electropuncture group. The VD model of mice was established by blocking the blood, tail bleeded and repeated cerebral ischemia- reperfusion. While the false surgical group is only a process. The blood wasn't blocked and the tail wasn't bleeded. They were treated on the operation day after they were waken. The shenshu, geshu, baihui were choosed on the electropuncture group, acupuncted with filifor,connected with electroacupunctrue instument,continuous waves,3 times/s. Ten minutes every time and one time a day. Xidezhen 0.77mg/kg/d were given to the xidezhen group through the means of watering stomach. The stoke index score and neural disorder were analyzed on the first and seventh day after the operation. Examined the study score through the water-maze experiment and the diving plat experiment on the seventh day after the operation and the memory score on the next day.Part2 The effect of electropuncture on hippocampus and cortical neuron apoptosia of cerebral ischemia reperfusion injury miceThe dividing groups, establishing models and treating methods were all same to part 1.3 mice were randomly choosed from each group on the first and seventh day after treatment.The tissue from optic chiasm to nipple was taken,fixated in 4% formal dehyde,covered with paraffin,maken into coronary slice(5um).One was choosed by every two slices. The cellular apoptosis on hippocampus and cortical neuron were observed by TUNEL.Part3. The effect of electropuncture on the cerebral cortex with Fas,Bcl-xL,P53 of cerebral ischemia reperfusion injury miceThe dividing groups, establishing models and treating methods were all same to part 1. The brain material was taken on the first and seventh day after treatment, broken away the bilateral cerebral cortex,fixated with 70% ethanol,prepared the single cell suspicions; the expressions of Fas,Bcl-xL, P53 were observed with flow cytometry.Statistical analysis of all data was conducted with SPSS 11.5。 Results:Part1. The effect of electropuncture on Praxiology of cerebral ischemia reperfusion injury mice1 the result of Praxiology scoreThe stoke index score and neural disorder in the model group were obvious high than the false surgical operation group on the first day after operation. The treatment of electropuncture can decrease the Praxiology score (P<0.01), Xidezhen has no effect on that (P>0.05).The two Praxiology scores of each group mice were similar to the false surgical operation group on the seventh day after operation and recovered from the motor disturbance.1 the results of study and memory scoreThe result of the study score and memory score of mice in the water-maze experiment were similar, the swimming time of the whole Line of the model group extended obviously and the mistake number of times increase singnifically (P<0.01), electropuncture and treatment with Xidezhen can decrease the swimming time of the whole Line and the mistake number of times(P<0.01), but the electropuncture was best than others(P<0.01).The result of the study score and memory score of mice in the diving plat experiment were similar, The react time of the model group extended obviously and memory latent period shorten (P<0.01), electropuncture and treatment with Xidezhen can shorten the react time and extended memory latent period (P<0.01), and the electropuncture was best than others (P<0.01).The result of the study score and memory score of mice in the electricmaze experiment were similar, the react time of the model group extended obviously and the mistake number of times increase singnifically (P<0.01), electropuncture and treatment with Xidezhen can decrease the react time and the mistake number of times(P<0.01), but the electropuncture was best than others(P<0.01).Part2 The effect of electropuncture on hippocampus and cortical neuron apopotosis of cerebral ischemia reperfusion injury miceApoptosis cells in hippo-CA1 zone and cortical of the false surgical operation group has no positive apoptosis cells.There were positive apoposis cells in the model group on the first day after operation,contraction into some cellular nuclear,khaki,nerve fiber disarray;positive apoptosis cells in hippo-CA1 zone were increased on the seventh day after operation,multi-apoptosis cells in the cortical,cortex became thin,a part flowing in hippo-CA zone neural cell,arranged in plexiform,lymphocytic infiltration,glial cell proliferation,focus of infaract could been seen;there was a few positive apoptosis cells in the Xidezhen group and electropuncture group.Part3. The effect of electropuncture on the cerebral cortex with Fas,Bcl-xL,P53 of cerebral ischemia reperfusion injury miceBcl-xL in cerebral cortex of the model group was decreased on the first day after operation (P<0.05),and was decreased obviously on the seventh day after operation (P<0.01). Treatment with electropuncture and Xidezhen on the first and seventh day after operation, Bcl-xL in cerebral cortex was increased (P<0.01 ),and electropuncture was better (P<0.01).Fas P53 in cerebral cortex of the model group was increased obviously on the first day after operation (P<0.01),and was increased obviously on the seventh day after operation (P<0.01). Treatment with electropuncture and Xidezhen on the first and seventh day after operation, Fas P53 in cerebral cortex was decreased (P<0.01), and electropuncture was best than others (P<0.01).Conclusion:The treatment with electropuncture (acupuncted the points of shenshu, geshu, and baihui) could up regulate the expressions of Bcl-xL, lower the expressions of Fas P53, inhibit the apoptosis of cerebral cortex neurons cells, strength the resist-damage and repairable ability, so it can improve study memory obstacle induced by Ischemia-Reperfusion in brain.
