Purpose: Take Radix puerariae thomsonii as research subject in order to establish a technical platform for traditional Chinese medicine extracted by bionic extraction method.Method: To evaluate the quality of ultra-powder by methods such as external appearance analysis, liquidity determination, determination of extractives, microscopy identification, TLC and content determination through contrastive Studies. To adopt single factor and orthogonal test optomizing the bionic extraction process. Use resin to purify the exreactive. Then establish the quality standard of the extractive. Preliminary study the metabolic way of the extractive through HPLC and HPLC-MS. Build the model of pharmacology to observe the antifebrile and analgesic pharmacodynamic effects.Result: The particle size of ultra-powder is smaller and distributed evenly, the cell wall almost completely broken, the content of extractive increased. The results of TLC and HPLC both released that the content of Puerarin and Pueraria Flavonid were significantly increased. The optimal process conditions of bionic extraction were determined through single factor and the orthogonal test. The solvent quantity was 100 times, temperature was 37℃, the agitation speed was 180r/min and mixing time was 20 mins. The content of Puerarin and Pueraria Flavonid was 0.7699% and 1.6232% separately. The type of macroporous adsorption resin was SP-70 that selected by static adsorption and desorption test. The optimal purification process as follows. The sample concentration was 0.5mg/ml, the volume of loading solution was 10BV, the velocity of flow was 2BV/h. The optimal elution process as follows. The impurity was washed by 6BV water and then washed by 3BV70% ethanol at the speed of 2BV/h. The quality standard is reliable, stable and with good reproducibility. The stability test in Vitro indicated that the extractive in artificial gastric juice and artificial intestinal juice were stable and no new composition founded. The biological samples such as urine, plasma, feces, heart, liver, spleen, kidney, brain, intestine, stomach were analyzed by HPLC qualitative analysis before and after given water solution of the extractive by means of imitate the course of oral administration, but only found 7 metabolites in the stomach and others in the form of prototype. The extractive can reduce the temperature that were induced by injecting dry yeast subcutaneously. The extractive could improve the pain threshold of mouses on the hot-plate and reduce the body twisting times induced by acetic acid in mouse.Conclusion: After the Radix puerariae thomsonii broken up through ultrafine grinding, the degree of uniformity significantly improved. After extracted by bionic extraction, the contents of Marker Component were improved vastly. The type of SP-70 resin has good purification effect, simple, convinent and feasible, can meet the mass production. The quality standard of bionic extractive can control its quality. The extractive mainly metabolized in the stomach and absorbed by the intestine in the form of prototype, spread widely. The culture in vitro and the metabolism in vivo are different. The extractive has apparent antipyretic and analgesic effects.
