Bladder Cancer Cell Proliferation And Smad4 Gene Expression Of Beta-elemene

Objective: To study theβ-elemene on cell proliferation in T24 bladder tumor cells and its expression of Smad-4 ,deeply investigate how theβ-elemene induced apoptosis of bladder cancer and its possible mechanism .In order to provide experimental evidence for the further study ofβ- elemene anti-cancer action and clinical application.Methods: T24 cells which are cultured in vitro, are affected by theβ-elemene on the consistency of 0.004mg/ml,0.008mg/ml, 0.01mg/ml,0.02mg/ml,0.04mg/ml,0.06mg/ml,0.08mg/ml for 24 hours, 48 hours and 72 hours later, The samples were detected by the MTT-method so as to show the effect ofβ-elemene on proliferation of bladder cancer T24 cells; and then use the software to calculate the consistence higher than the minimum IC50. Use the Western-blot , RT-PCR assay and cell morphology to reveal the influence on the expression of Smad-4 protein and the level of mRNA ,and also show bladder cancer cells morphological changes.The comparative group(ie.0mg/ml) is set up for comparation.Results: MTT assay results showed that:the minimumβ-elemene concentration which can inhibit more than 50% T24 cell was 0.02mg/ml; while the other treatment groups of drugs can inhibit the growth of T24 cells and the survival rate of T24 cells are reduced with the concentrations increased or the time extended.It means that dependence of dose and time existed.Compared with the comparative group this result were statistically significant (p <0.05), Compared with every two treatment group shows that it was statistically significant (p <0.05).the Western-blot and RT-PCR result of T24 which were affected under the final cancentration of 0.02mg/mlβ-elemene for 24 hours, 48 hours and 72 hours shows:β-elemene can enhance Smad-4 protein in T24 cells and The expression of mRNA and time-dependent manner; T24 cell morphology showed that: with the time, 0.02mg / m concentration ofβ-elemene treatment group, cell shape gradually become round or irregular , the number increased adherence capacity decreased, the surface of sprouting, cell volume was significantly reduced, plasma concentration, the nucleus was significantly smaller proportion appeared floating single cells and caused some cell disintegration into pieces,or loss of normal cells form.Conclusion:β-elemene could inhibit the T24 bladder cancer cell growth and proliferation, which may enhance the T24 cells with Smad-4 protein and mRNA expression...
