Objective: To observe the effects of Kaiyuqingre's Prescription on the glucose and lipid metabolism,and the expression of Monocyte Chemoattractant Protein-1(MCP-1) in adipose tissue of spontaneous Type 2 Diabetes Mellitus rats(OLETF rats).Method: A control study was carried out between the spontaneous Type 2 Diabetes Mellitus rat(sOLETF rats)and LETO rats,and all OLETF rats were divided into three groups randomly:Model group,Metformin group and Kaiyuqingre's Prescription group.Blood glucose,lipid and insulin resistance related indexes were determined.Immunohistochemical method and real-time flourescent quantitative polymerase chain reaction(PCR)technology were used to detect the expression of Monocyte Chemoattractant Protein-1 in adipose tissue from the protein and gene levels in each group.Results: 1,Contrast to the control group,there is a lower level of OLETF rats'blood glucose,total cholesterol,triglyceride,free fatty acid,body weight and intra-abdominal fat content,and a higher glucose infusion rate in the Kaiyuqingre's Prescription group.2,Expression of MCP-1 protein in rats'adipose tissue:â‘ The results of immunohistochemistry showed:MCP-1 expressed limiter and weaker in LETO rats'adipose cells.The Modal group's expression was obviously increased,and the Kaiyuqingre's Prescription group was weaker than model group.â‘¡Quantitative analysis showed:contrast to LETO group,positive staining cell area and mean optical density of the expression of MCP-1 in adipose tissue is higher in the Modal group.Contrast to the Modal group,the expression is lower in the Kaiyuqingre's Prescription group.3,The MCP-1 mRNA expression in rats'adipose tissue:contrast to LETO group,the Model group was significantly higher.Contrast to Modal group,the Kaiyuqingre's Prescription group is significantly lower.Conclusions: 1,Kaiyuqingre's Prescription has the effect of hypoglycemic action,lipid-regulating,weight reduction and improving insulin resisitance.2,The protein and gene of MCP-1 in OLETF rats'adipose tissue expressed higher,it suggested that MCP-1 may have an important role in pathogenesis of adipose tissue's micro-inflammation in the Type 2 Diabetes.Through the result of lower MCP-1 expression in the Kaiyuqingre's Prescription group,it revealed MCP-1 be the target of this chinese herbal compound.
