| Purpose:To solve the problem that longhairy antenoron herb preparation which applied on the market nowadays ,although has good effect,it has large dose once and inconvenience when taking; To solve the problem of counterfeit drug of longhairy antenoron herb prevailing which can harm to health.Method:①T o study the mobility,extract and moisture of submicron powder, carry on research by using semi-quantitative thin-layer and quantitative HPLC, comparing with regroundgrain,formulate the quality standards of submicron powder;②As an indicator of total flavonoids,explore macroporous,polyamide adsorption,ethyl acetate extraction,butamol extraction, water extraction and alcohol precipitation to purification longhairy antenoron herb differences,determine the best separation and purification methods,than examine the various factors affecting this method,the optimum conditions and parameters,enrichment as much longhairy antenoron herb active site-flavonoids to reduce the preparation single purpose of taking large doses;③U sing HPLC-MS method,establish genuine fingerprint of longhairy antenoron herb,make full control of money grass from the overall quality of medicines,solve the problem source confusion.Result:①To formulate the quality standards of submicron powder.Longhairy antenoron herb has light color and with clumps,worse mobility than the power fluidity,content and water extract were higher than in powder and moisture absorption than powder;Microscopic identification results show that superfine in microscopy showed irregular granular material filled with small vision;Thin semi-quantitative and quantitative comparative study of HPLC shows longhairy antenoron herb ultrafine active ingredient in the powder were higher than regroundgrain;②The results of the purified shows NKA-9 type macroporous resin separation and purification of total flavonoids money grass comprehensive adsorption separation performance than other types of macroporous resins,ultimately determine the NKA-9 type macroporous resin purification of total flavone of optimum conditions:the sample concentration 20mg/ml,to 6 BV the amount of distilled water,70% ethanol elution,after purification, the purity of 59.4% of total flavonoids;③Through the exploration and optimization to determine conditions:column: Shim-pack VP-ODS(4.6×250mm,5μm);mobile phase:acetonitrile-0.1%formic acid;flow velocity: 1ml/min;the detection wavelength: 370nm;column temperature: 30℃;the injection volume: 10μL;run 90 minutes.In the chromatographic conditions,the establishment of a common pattern longhairy antenoron herb,established the 10 peaks,10batches of medicines in different habitats peaks of the relative retention time and relative peak are smaller than 3.0% and the similarity between the 0.708~0.974.And through the reference substance and the MS diagram,is identified peak5,6,7,respectively,rutin,quercetin,kaempferol.Conclusion:This is the first on the longhairy antenoron herb powder superfine and the nature of a more systematic comparative study,found that when the longhairy antenoron herb made by the pulverization of the effective part has increased the content of total flavonoids;By NKA-9 type macroporous resin,flavonoids can effectively achieve the purpose of enrichment,which can reduce the single dose preparations; longhairy antenoron herb for the first time established a genuine fingerprint of HPLC-MS,identified 10 peaks and 10peaks were identified in the three peaks of the structure,and other common components need to be further identified through other means.This will help to effectively observe and compare different habitats longhairy antenoron herbal al ingredients contained in the proportional relationship between the content in order to enhance the effectiveness of different longhairy antenoron herbs origin evaluated the quality,the authenticity of identification for longhairy antenoron herbal. |
