Objective:To observe differences between Ephedra, Schisandra on the angiogenesis for treating rat idiopathic pulmonary fibrosis A5 induced by bleomycin.And explore the academic gist and functional mechanism of these differences.Method:(1) Document research:The paper summarizes the main progresses in the def inition, clinical presentation, etiology, mechanism, diagnosis and treatment of idiopathic pulmonary fibrosis. And we observed differences between ephedra, Schisandra on the angiogenesis of lung tissue of the idiopathic pulmonaryry fibrosis animal model,based the viewpoint that the angiogenesis is the fundamental pathological mechanism of the idiopathic pulmonary fibrosis, in order to provide the theory basis on the treatment of idiopathic pulmonary fibrosis (IPF). (2) Exper iment in vivo:①Establishment of the model rats of idiopathic pulmonary fibrosis;②From the changes of weight, skin, respiratory rate and the lung coefficient, we have obsreved the impact of Ephedra, Schisandrae on the generalstat state of rats;③By observing the pathophysiology of lung injury, measuring the inner diameter of pulmonary arterioles, density of the nuclear membrane, we have studied the impact of Ephedra, Schisandra on pathological injury of small artery;④By counting the number of rats lung tissue after treatment of Ephedra, Schisandra, we studied the condition that Ephedra, Schisandra on the angiogenesis for treating rat idiopathic pulmonary fibrosis;⑤By measuring the content of ET-1, and 6-Keto-PGF1a, which were measured by IRMA, we have studied the mechanism of Ephedra, Schisandra on the angiogenesis for treating rat idiopathic pulmonary fibrosis.Results:①General state:models established, the rats'carry momentum decreases, furgrown dark by degrees.and lost luster.The body weight of rat gain slowed down. The rate of increase was the slowest in the model group. The weight gain of the model group significantly droped behind the normal group (P< 0.05).The increase of respiratory frequency was significantly (P< 0.01). Ephedra,and Schisandra chinensis used in combination can significantly improvement the respiratory frequency of rats (P< 0.05). The lung coefficient of the model group increased and had significant difference between the normal group (P< 0.05).The respiratory frequency of the Ephedra group and the HC group was significant improved (P< 0.05).②The impact on the pulmonary artery injury:The alveolitis grades and IPF induced by bleomycin of the model group were sensible increased then the normal group (P<0.05). Both the Ephedra group and the HC group can significant restrain the alveolitis grades and IPF (P< 0.05). Content of ND and ED had significant differences between the model group and the normal group (P< 0.01).Content of ND and ED of the Ephedra group and the HC group was significantly improved (P< 0.05).③The angiogenesis of the arteries:Number of the blood vessel of the model group restrain was significantly more then the normal group (P<0.05); the Ephedra group was significantly weaker then the HC group in restrain the arteries of the lung(P<0.05). the number of the blood vessel ofthe Ephedra group is higher then the Schisandra chinensis group and Ephedra and Schisandra chinensis used in combination group.④The impact on related cytokines:at 7d, the level of ET-1 of the model group had significant difference with the normal group (P< 0.05),the level of ET-1 of the model group was at a higher level, and had more significant difference with the normal group (P< 0.01).The level of ET-1 of Ephedra and Schisandra combination used group and the HC group can be significant descended. At 7d, the level of 6-Keto-PGF1a of the model group was higher than the normal group,but there was no significant difference (P>0.05). There was the same condition between the model group and every treatment group. At 7d, the level of 6-Keto-PGF1a of the treatment group rised,and there was significant difference between Ephedra,and Schisandra combination used group and the normal group (P< 0.05)Conclusions:theoretical study and experimental results showed that:①"Ascent-descent disorder of visceral qi " is the basic pathogenesis of IPF.②Ephedra has positive effect on restrain the increase of respiratory frequency. Ephedra,and Schisandra chinensis used in combination can significantly restrain the impact on the pulmonary artery injury; Ephedra,and Schisandra chinensis used in combination can restrain the angiogenesis of the arteries by reducing the level of ET-1 and promoting the level of 6-Keto-PGF1a in blood.③the degree of Ephedra, Schisandra on the academic of the angiogenesis is different, but this influence to the angiogenesis is the main step of the influence to the IPF.
