Limitations With Locally Advanced, Metastatic Renal Cell Carcinoma Difference In Serum Protein Expression

Objective:Surface enhanced laser desorption/ionization time of flight mass spectrometry (SELDI-TOF-MS) is a useful technique in finding the changes in protein expression of different staging of renal cell carcinoma(RCC). In this study, we screened the differential expressed protein in the serum of localized RCC,locally advanced RCC and metastatic RCC patients and explored the relation between the differential expressed protein and advanced renal cell carcinoma. This will provide us a method for staging of renal carcinoma in protein level. Methods:31 serum samples of renal cell carcinoma patients were used for the screening of differential expression protein by SELDI-TOF-MS with CM10 protein chip.18 of the 31 samples were localized RCC (Ⅰ+Ⅱ) patients, the rest were locally advanced RCC and metastatic RCC (III+IV) patients.Results:6 peaks were detected in the molecular weight ranging from 2.0 X 103-20X103 (2.0-20KD), in which 5 (2674.47,11678.93,6018.73,3395.03,3483.54) were expressed higher in locally advanced RCC and metastatic RCC than in localized RCC and 1 was lower in in locally advanced RCC and metastatic RCC than in localized RCC. The P value is lower than 0.01.Conclusions:6 proteins expressed differentially between localized RCC and advanced RCC.5 of 6 differentially expressed proteins expressed higher in advanced RCC than in localized RCC which indicates that the expression of these differentially expressed proteins are positive correlated with the progression of renal carcinoma. One differentially expressed protein (6885.27) expressed lower in advanced RCC than in localized RCC. These results will provide us a new method in the staging of renal carcinoma.
