[Objective]:We study the effect of Pseudomonas Aeruginosa Injection (PA-MSHA) treatment on cell proliferation, apoptosis, migration and invasion on mouse pancreas cancer cell line (MPC).[Methods]:We used MTT assay to measure cell viability of MPC at different PA-MSHA concentrations and treatment intervals, therefore determined the optimum PA-MSHA treatment effectiveness. Under the similar experimental conditions, we also examine the cell morphological changes using microscope. We performed Hoechst33258 nucleus staining assay to observe apoptotic cells under the florescence microscope. We used flow cytometry to analyze MPC cell cycle modifications under PA-MSHA treatment. We used cell scratch test and transwell assays to investigate cell migration and invasion.[Results]:Under in vitro condition,we observed more than 95% MPC cells alive. The cells reached the logarithmic grow phase after 12 hour of culturing.The MTT test showed time dose dependent effect of the PA-MSHA.Under the florescence microscope,we saw clear evident of cell apoptosis.This is supported by flow cytometry assay, which showed a typical apoptotic peak.When the PA-MSHA concentration reaches 2~6×108 cfu/ml, the cells in G2/M cell cycle phase increased, which indicated that the treatment could induce G2/M cell cycle phase arrest.Finally, the transwell and cell scratch test showed that the PA-MSHA could prohibit cell migration and invasion.[Conlusion]:Our study showed that PA-MSHA treatment can prohibit MPC cell proliferation in a does time dependent manner.We also showed that the PA-MSHA could induce cell apoptosis,and reduce cell migration and invasion.Therefore,we postulate that the PA-MSHA treatment has potential in treating pancreas cancer.
