| Objective To observing the affection on the morphocytology of podocyte,the expression of TGF-B and VEGF,in the DN rats'renal tissue who were treated by Tripterygium wilfordii astragaloside mixture.Method Use the high-fat high-sugar diet to feed the SD male rats for one month,after intraperitoneal injection of STZ rats,resulting in DN rat model,and forming rats were randomly divided into Model group,Losartan group,Tripterygium group,low,medium and high does Tripterygium wilfordii astragaloside mixture group and normal group(non-model), at 4th,8th,12th,weeks,collect the blood and unine specimens,test the glucose(GLU),triglycerides(TG),cholesterol(TCH),serum creatinine(SCR),alanine aminotransferase (ALT) ang 24 hours of quantitative protein level,and killed all rats at 12th weeks,colleted renal tissue specimens and calculated kidney weight index and to use immunohistochemical chemical method to observe rat kidney tissues for TGF-βand VEGF expression,as well as electron microscopy of Change podocyte.Result Astragalus tripterygium mixture,the medium,high dose group of rats 24h quantitative protein levels were significantly lower than the model group,losartan group and tripterygium Decoction Group;ALT values,tripterygium decoction group and mixture group was significantly lower than the model group;lipid values,the treatment group were significantly lower than the model group;rat renal tissue by immunohistochemistry showing: DN rat renal tissue TGF-β,VEGF expression increased in normal rats;but Losartan group, tripterygium decoction group were weaker than model group,and the mixture group was significantly weaker than the two groups;rat renal tissue showing electron microscopy:DN rats have a different degree of foot process fusion,mixture group was significantly less than the model group,losartan group and tripterygium decoction group,and the low,middle and high dose group had no difference.Conclusion Tripterygium wilfordii astragaloside mixture can not only significantly inhibited TGF-β,VEGF in renal tissues,which play a protective effect of podocyte,also regulate lipid metabolism DN rats,the protection of liver function,lower albumin excretion rate. |
