Ramoplanin is one of the compounds under clinical researching used to treat the MRSA (Methicillin resistance Staphylococcus aureus) and VRE (Vancomycin resistant Enterococcus ).infectionsA strain (Actinoplanes sp.), SIPI-A.2001, was used in this study. First, strain improvement was made by natural selection andthe fermentation optimization was also made. The results of single factor experiments showed that bean oil and glucose were the best carbon sources, soybean powder was the best nitrogen source. Filling 100ml medium in a 750ml shake flask was suitable, the best inoculation ratio was 10%, the best initial pH of fermentation medium was 7.0, suitable cultural temperature was 25℃. On the other hand, feeding L-Leu into the broth before cultivation increased the fermentation potency. Compared to that of basal condition without any addition of precursor, the productivity was increased to 150%.The purification process of ramoplanin was that after pre-prcessing the broth, using macroporous ion exchange resin to purify the product and using Al2O3 to remove the pigment..Finalythree purified ingredients were obtained and there structures were identified by the analyses of their physico-chemical properties,MS ,UV and IR spectra data. which were identical with ramoplanin A1,ramoplanin A2 and ramoplanin A3 reported.The gene of orf15 encoding cyclase of ramoplanin biosynthesis .wascloned and its genetic transfer system was also build up.
