Purpose: To study the structure changes of cutaneous tissue of intrinsic aging mice received the extraction of Astragali by intragastric administration. To investigate the anti-aging effect and degree of Astragali. To discuss the anti-aging mechanism of Astragali by Morphological way. In order to define and demonstrate the role of anti-aging and its clinical significance of Chinese traditional medicine.Material and method: Ten 4-month-old mices were in forming a group as youth (A). Fifty 18-month-old mices were randomly divided into 5 groups, 10 micess per group: aged blank group (B), low dose group (C), middle dose group (D), high dose group (E) and VitE-C matched group (F) . All of them are health female Kunming kind. Gave the group A, B NS 20ml/kg by intragastric administration accordingly, the group C, D, E the extraction of Astragali 4g/kg, 8g/kg, 16g/kg, in antother word 20ml/kg drug every time by intragastric administration accordingly, the group F VitE-C suspension 20ml/kg by intragastric administration. All above was handled 1 time per day. After 30 days , cut the hair on the back of mices, about 2cm×3 cm. Then killed the mices and cut the decaying skin 1 cm×1 cm. After fixing and making section, the skin are studied.Results:1. Comparing with group A, the epidermis thickness(P<0. 01, respectively), the counting of fibroblast (P<0. 01, P<0. 01, P<0. 01, P<0. 05, P<0. 01, respectively) , the area of collagenous fibers(P<0. 01, P<0. 01, P<0. 05, P<0. 05, P<0. 01, respectively) of all the other groups are decreased.2. Comparing with group B, the epidermis thickness (P<0. 05, P<0. 01, P<0. 01, P<0. 01, respectively) , the counting of fibroblast (P<0. 01, respectively) , the area of collagenous fibers (P<0. 01, respectively) of group C, D, E, F are increased. 3. Comparing with group D, E, F, there is no statistical difference between group D and E, nor group E and F at the aspect of the epidermis thickness, the counting of fibroblast and the area of collagenous fibers. While, comparing with group D and F, the epidermis thickness (P<0.05, respectively), the counting of fibroblast (P<0.05, respectively) and the area of collagenous fibers (P<0.05, respectively) are increased.Conclusion:1. The epidermis thickness, the counting of fibroblast, the area of collagenous fiber are decreased in intrinsic aging mice.2. The epidermis thickness, the counting of fibroblast, the area of collagenous fibers are increased after giving the extraction of Astragali by intragastric administration. The result justifies the definite function of Astragali to postpone senility. This function may be more obvious used high dose drug.
