Objective:To study the therapeutical effect of Oleanolic acid(OA) and its mechanisms on transplanted CML mice models, to provide experiment for clinical application of OA.Methods:Leukemia murine models were established by injecting K562 cells into severe combined immune-deficient(SCID) mice via abdominal cavity.Various methods,such as blood film,bone marrow graph,WBC and histopathology,were applied to detect the leukemia cells infiltrated in the peripheral blood,bone marrow,liver and spleen of CML models, And the expression of CyclinD1,CyclinA,Bcl-2 in liver and spleen were detected by immunocytochemistry.Results: After injection of K562 cells for 4 weeks, leukemia cells had been found in the peripheral blood,bone marrow,liver and spleen of SCID mice.OA can improve the general states of models, increase mice's weight,prolong their lives,decrease the number of leukemia cells in the peripheral blood,improve blood and bone marrow,and can inhibite the expression of Bcl-2,CyclinD1,CyclinA in leukemia cells infiltrated in liver and spleen.Conclusion:1. OA can significantly improve the symptoms of CML models, indicating that a certain effect on the CML models.2.The expression of Bcl-2,CyclinD1,CyclinA in leukemia cells infiltrated in liver and spleen can be inhibited by OA.
