| Tiarella. polyphylla D. Don. belongs to the family Saxifragaceae recorded by "The medical plants of Sichuan Wulong" firstly, The whole plant was widely utilized as folk medicine in China, which was confirmed as cold-natured, bitter in taste, asepsis, and focus on heart, liver and stomach. It has efficiencies of heat-clearing and detoxicating, promoting blood flow, detumescence and alleviating pain, and it is effective to cough and asthma, injury from fall, hepatitis and so on. Tiarella. polyphylla D. Don. was traditionally used as an important folk medicine, with the abundant resources in east Asia, however, there are rarely reports on it's main active components and pharmaceutical research. Therefore, researches on material foundation of medicinal effectiveness and quantitative analysis of active components from Tiarella. polyphylla D. Don. were conducted.In this thesis, the chemical constituents of the extract of 85% ethanol from the whole plant of Tiarella. polyphylla D. Don. were studied, 9 compounds were isolated on columm of silica gel, polyamide and pured with TLC, recryst etc. On the basis of analysis of spectroscopic data and measuring of physico-chemical characters, the chemical structure of 7 compounds were confirmed as stigmasterol (1), heptadecanoic acid (2), ethyl 3,4,5-trihydroxybenzoate (3), quercetin (4), protocatechuic acid (5), myricetin (6) and gallic acid (7). The structure determination of the two other compounds will be finished later. Compound 1, 2, 3, 5, 7 were isolated for the first time from Tiarella. polyphylla D. Don.Active components in Tiarella.polyphylla D. Don.such as triterpoids and flavanoids compounds were quantitative analyzed by RP-HPLC with the index of oleanolic acid, myricetin, quercetin and kaempferide, both of the methods are accurate , repeatable with a good stability. Furthermore, NIR method was applied in the determination of quercetin in Tiarella. polyphylla D. Don., the predictive value by the method we have established was in the range of relative accuracy, which indicated that NIR is suitable for the determination of flavonoid compound with low content. The research on the quantitative analysis will provide basis for quality control evaluation of Tiarella. polyphylla D. Don.The method of water-steam distillation was used to exact the essential oils from three parts (root, stem, leaf) of Tiarella. polyphylla D. Don., the qualitation and quantitative analysis were conducted with GC-MS combined with the stoichiometry method (Heuristic Evolving Latent Projections method), there are some common components such as linalool,α-terpineol, laurostearic acid, hexadecanoic acid, phytol and linoleic acid in three different parts of Tiarella. polyphylla D. Don.The pharmacal action corresponding to the active components isolated from Tiarella. polyphylla D. Don. and the classify to the plant from the point of photochemistry will be explained by the result of this thesis, which will also provide refered value on developing and modernization of natural medical resources in our mather land. |
