Objective:Explore the protection of Wuji Solution (He Ji Ju Fang) on when Helicobacter pylori(H-pylori) infect mouse gastric epithelial cells(C57BL/6 strain); analyze such protection on both the apoptosis of gastric epithelial cells in mouse and the resisting of H·pylori in mouse body.We examine the impact of Wuji Solution to the cytokine IL-1β,IL-8 and IFN-γ's content in inoculated mouse serum, in order to grant further evidences on Chinese medicine therapy on H.pylori gastritis.Methods:Use H.pylori inoculate C57BL/6 strain mouse, and feed the strain with medicine for 7 days. In the purpose of observering the protection role of Wuji Solution on gastric epithelial cells, and the fuction of the medicine in killing H.pylori; we examine inoculated mouse by HE dying, fast urease test and bacteria cultivation the body in strain mouse; Use flow cytometry analyze the extent of apoptosis of gastric epithelial cells of inoculated mouse. Use. enzyme-linked immunosorbent assay (ELISA) to elucidate how Wuji Solution influence the infection of H.pylori to the cytokin IL-1β,IL-8 and IFN-γin the serum of inoculated mouse.Results:(1)Compared with model group,the pathological change in each treatment groups have enormous difference (P<0.05), each dosages of Wuji Solution could reduce thepathological change in gastric tissue(2)The number of each dasage of Wuji solution has enormous difference with model group colony(P<0.05). On contrast of model group, colonies in high dosage of Wuji solution do not have such differece with clarithromycin group. Thus, Wuji solution can surpress H.pylori in mouse.(3) The fast urease test has differences between every group (P>0.05), fast urease test result has difference among groups between every group. With rising of dosage, Wuji Solution antibacterial result of liquid strengthen。(4) The apoptosis of cells in Wuji Solution group is lower than that of model group (P< 0.05),in the mean time, each treatment group of apoptosis is descend. Colonies in Wuji solution do not have obvious differece with clarithromycin group.(5) The content of mouse serum (IL-1β) in model group increases (P<0.05) notably than normal group; each treatment group serum has low content IL-1βin contrast with model group. Both the content of IL-1βin Wuji Solution group and clarithromycin group do not have different from that in normal group's serum (P> 0.05).(6) The content of mouse serum (IL-8) in model group increases (P<0.05) notably than normal group; each treatment group serum has low content IL-8 in contrast with model group. Both the content of IL-8 in Wuji Solution group and clarithromycin group do not have different from that in normal group's serum (P> 0.05).(7) The content of mouse serum (IFN-y) in model group increases (P< 0.05) notably than normal group; each treatment group serum has low content IFN-yin contrast with model group. Both the content of IFN-yin Wuji Solution group and clarithromycin group do not have different from that in normal group's serum (P> 0.05).Conclution:(1)Wuji Solution has bacteriostatic effect in inoculated mouse, increase the percent of turning negative in urease reaction. Wuji Solution can protect gastric tissue,and can inhibite the apoptosis of gastric epithelial cells.(2) Wuji Solution inhibites the production of IL-8, IFN-γ, IL-1β,and reduces inflammation on inoculated mouse organisms.
