Multidrug Resistance Schisandra Soft Capsule Pharmaceutical Research

Objective: To study the pharmacology material base of soft capsule of Fructus Chisandrae on the reversal effect on multidurg resistance, in order to lay a theory foundation of development and application in clinical.Methods: First, according to the active ingredients of Fructus Chisandrae, determined the dosage form preparation, extraction method. We used orthogonal experimental method to determine the optimum extraction process. Second, we initially tested the efficacious forms of Fructus Chisandrae on pharmacology, including the study of the extraction of Fructus Chisandrae of the inhibition of transplanting tumor and the study on the reversal effect of multidurg resistance of KBv200 and MCF-7/Adr tumor cells, which acquired resistance. And we preparated the forming process of soft capsule of Fructus Chisandrae. At last, we identified the soft capsule of Fructus Chisandrae by TLC chromatography, and adopted schisadrin as the index by high performance liquid to test the content of the dosage.Results and conclusions: Soft capsule of Fructus Chisandrae for the technological process for the final: Fructus Chisandrae, immersed in water, peeled the pulp off, dry the seed, crushed into powder, six times in 90% ethanol to extract three times, followed by 2,2,1 hours, filter, and the filtrate decompression recovery ethanol, drying, crushed into powder(100 head); Mingled 5% edible soya-bean oil and 0.5% beeswax, heating to 80℃so that the beeswax dissolved completely, cooling below 35℃, mixed with dry cream powder, grinded well-distributed, lay aside for 3 hours, exhausted air to vacuum and clear up bubble, pressed soft capsule by machine, de-fat by 95% ethanol, drying below 30℃for 60 hours, to the degree that the surface of capsule was dry, and had appropriate hardness; With pharmacology experiments, it proved that the extraction of Fructus Chisandrae in different dose could inhibit the growth of transplanting tumor to different degree, prolong the lives of animals that had tumor, and reverse multidury resistance of KBv200 tumor cells to VCR and MCF-7/Adr tumor cells to DOX with dependent dose; quantitatively analyzed the interrelated main component part with advanced measure technique of chromatography, in order to establish definite index of the control standard of quality and guarantee its steady.
