Objectives:According to the traditional ethnic medicine theories and the clinical background of Mongolian drug Agi, to study the effects of drug-processing on Agi chemical compositions and hemostatic effects, and to discuss some of the hemostatic mechanism of raw Agi and charred Agi.Methods:1. The optimized processing procedure was screened by orthogonal test according to characters, the clotting time and the content of the extraction.2. The species of flavonoids in samples were identified by color reactions, and the contents of total flavonoids were measured through UV spectroscopy using rutin as control.3. Inorganic elements (K, Ca, Mg, Zn, Fe, Mn, Cu, and Al) contents in raw Agi and charred Agi were determined by ICP-AES.4. The mouse bleeding time was measured by tail breaking test, and the clotting time was measured by capillary test.5. The rabbit bleeding time was measured by traumatic hemorrhage test, and the clotting time was measured by tube test.6. The rabbit prothrombin time (PT), activated partial thormboplastia time (APTT), thrombin time (TT), fibrinogen (FIB), plasma recalcification time (PRT), euglobulin lysis time (ELT), max platelet aggregation rate (MPAR) were measured by solidification method, turbidimetry and tube test to ananlyze the effects of raw Agi and charred Agi on rabbit coagulation-fibrinolysis systemn and platelet function.Results:1. The temperature has the crucial efflucences on the drug's quality (P<0.05), and the results of analysis of range manifest showed that the charring time can also affect the quality of the drug in certain degree.2. The main components of the flavonoids in raw Agi are flavone and flavonol, while the main components of cherred Agi are chalkone and isoflavone. The average contents of total flavonoids in raw and charred Agi were 4.78% and 2.26% respectively.3. The sequences of inorganic elements contained in the raw Agi and charred Agi are K > Ca > Mg > Al > Fe > Mn > Zn > Cu. After drug-processing, the inorganic elements were increased on different degree, but dissolution rates were decreased apparently in water.4. Effects of Agi on the mouse BT and CT: In the group with medium doses and high doses of charred Agi, BT was significantly declined (P<0.01); the high doses of raw Agi group, medium and high doses of charred Agi groups manifestly shortened mouse CT (P<0.01).5. Effects of Agi on the heparinized mouse BT and CT: both raw Agi and charred Agi decurtated heparinized mousse BT and CT significantly (P<0.01).6. Effects of Agi on the rabbit BT and CT: the medium doses and high doses of raw Agi groups, the low, medium and high doses of charred Agi groups decreased rabbit BT obviously (P<0.01 or P<0.05); all of the groups of raw and charred Agi declined rabbits CT (P<0.01 or P<0.05).7. Effects of Agi on rabbit coagulation systemn: all of the doses groups of raw Agi and charred Agi have no apparent influences on PT (P>0.05); the high doses of raw Agi group and the low, medium and high doses of charred Agi groups decreased APTT apparently (P<0.01 or P<0.05); the high doses groups of raw Agi and charred Agi still decreased TT apparently (P<0.01 or P<0.05); the medium and high doses groups of charred Agi increased FIB obviously.( P<0.01 or P<0.05).;the high doses group of charred Agi showed the decreased PRT significantly (PO.05).8. Effects of Agi on rabbit fibrinolysis systemn and platelet function: the high doses group of raw Agi groups, the low, medium and high doses charred Agi groups prolonged ELT (P<0.01 OR P<0.05); the high doses group of charred Agi increased MPAR obviously (P<0.01).Conclusions:1. The optimized processing procedure of charred Agi was as follows: 210℃for 20 minutes. The screened procedure is a fast and convenient method suitable for the control of the processing technique of Agi.2. Flavonoids are the main chemical composition of Agi. Drug-processing had some effect on the species and the content of total flavonoids.3. The inorganic elements which are essential to hemostatic effects are rich in Agi, and all the analyzed elements contents were increased after drug-processing. In contrast, the inorganic elements extraction rates declined significantly.4. The raw Agi do has the favorable effects on haemostasis and coagulation, and these effects were inhanced after drug-processing.5. Raw Agi can play its role in haemostasis and coagulation by affecting the intrinsic pathway of coagulation and fibrinolytic system. These effects were inhanced after processing drugs; moreover, the charred Agi could increase FIB and MPAR, with promoting more in blood coagulation.
