Subject:To find a new stratagem for developing of Chinese traditional medicine by the evaluation the anti-infectious bronchitis virus effect of the micronized Indigotica FortMethods:Prepared the micronized Indigotica Fort powder by dry grinding method and verified the diameter of the powder by transmission electron microscopy firstly. Secondly, studied the toxin effect of the micronized Indigotica Fort powder by MTT method on the primitive chicken renal cell from the chicken embryos to find the proper concentration for the research. In vitro study, we evaluated the anti-IB V effect of the different diameters of the Indigotica Fort powder through indirect immunofluorescence and PCR method to expound the relation between the diameter of the powder and the anti-virus efficacy. In the vivo study, we toke the chicken embryos and the1-day-old chicks as model in the research. Observed the death peak, survival rate, pathological degrees, clinical symptoms and the IFN-y level in the serum of the chicken embryos and the1-day-old chicks by affected infectious bronchitis virus(type M41, MOI=5), compared the difference between the group treated with micronized Inidigotica Fort powder and the group treated with untreated Inidigotica Fort power to find the influence on the Indigotica Fort biological efficacy by micronization. Results:It was successful acquisition the different diameter of Indigotica Fort powder by dry grinding method. The diameter of550μm,250μm and100μm were in integrated structure by transmission electron miscroscopy. In the toxic effect of the micronized Indigotica Fort powder in vitro, we found that the micronaztion procession could reduce the toxic effect of the Indigotica Fort.In further study, we found that the anti-virus effect of the micronized Indigotica Fort increasing following by the reduce of the diameter.The diameter of100μm was the best proper size for the Indigotica Fort to show the best anti-viral efficacy. In the vivo study,compared with untreated Indigotica Fort group, the micronized Indigotica Fort group had much better anti-virus effects on prolong the death peak, increasing the survival rate of the chicken embryos/chicks, relief the degrees of pathological and the clinical symptoms,but there was no change of the IFN-r level in serum of the chick affected by infectious bronchitis virus(MOI=5).Conclusion:It was simple to process,lower cost and easy to operate the micronized Indigotica Fort powder by dry grinding method which showed much feasible of developing Chinese traditional medicine. Secondly, the micronization had much efficacy such as reduce the toxin of the medicine,enhanced the anti-virus effect and so on.But one interesting thing was that the IFN-r was not change in the micronized Indigotica Fort group, which opposite our primary suppose. So we suppsed that the micronization would cause the mechanism change of the Indigotica Fort group.
