| The major activity components of Uncaria are Rhynchophylline and Isorhynchophylline.And the paper used them as effective fractions to do the research to authentication therapeutic effect to ischemic stroke animal model induced by MCAO with suture method of Uncaria alkaloids,and chemical composition analysis. The thesis include six parts as follow:1.The manufacture ischemic stroke animal model induced by MCAO with suture method.2.Therapeutic effect to ischemic stroke animal model induced by MCAO with suture method of Uncaria alkaloids.And Have a therapeutic effect.3.The preparative technics of effective fractions have been researched.Viaing absorption by macroporous resin,determine the optimum extraction and purification process,that's 8 times 80%ethanol,extract 3 times and 2 hours each times.4.Preparation of reference substance,apply Silica gel(sanple:silica=1:200) column chromatography obtain two reference substance.5.Investigation of method for quality control of effective fractions.The contents of the Rhynchophylline and Isorhynchophylline in the effective fracions were determined by UV spectrophotometer,and HPLC method was established to determine the ether extraction contents of Rhynchophylline and Isorhynchophylline. |
