Rodin Lipid Capsules Pre-clinical Pharmacy And Pharmacodynamic Study

Luodantiaozhi capsual, developed from the edible items of the Department of Health definition, is used for the people of hyperlipemia and indisposition.Modern purification technology was applied to extract the essential components from the Chinese herbs, such as Apocynum venetum L.,Flos chrysanthemi, Radix et rhizome salviae miltiorrhizae,Fructus crataegi,Fructus lycii,Rhizoma gastrodiae.The capsual fully showed the characteristic of multicomponent,multistrata,multitarget.In this article the pre-clinical pharmaceutical,pharmacological tests of the capsual were studied,which laid the foundation to the further development.The results of study were as follows:1. The purification method was established through Pharmacopeia, pre-processing and active ingredients.2. The best composition of preparation and the optimization of processing technology were selected by orthogonal experiment.The optimized process was as follows: the part of alcohol extraction, 60% ethanol, 10 times the volume of raw material, extracted twice, each time 1 hour; then, the part of water extraction, 8 times the volume of raw material, extracted twice, each time 1 hour; The process is simple and easy, which laid foundation for the industrial production.3. Pilot study and capsual forming process were carried out. The results revealed that the technology was applicable to industrial mass production, the best forming process: Microcrystalline cellulose: Dextrin (1:1).4. An HPLC method was established to determine the content of Gastrodin, and an UV method was used to determine the content of the total flavonoids and the polysaccharides.The methods were reliable, accurate and could be used for the quality of the capsual.5. The acceleration stability and long-time stability experiments of capsual were carried out. The results of current three months showed the capsual was steady under 40℃high temperature, 75% humidity and normal temperature with 60% humidities conditions. The characteristics didn′t appear obvious variety. The contents of the Gastrodin, the total polysaccharides and the total flavonoids met the requirements. The capsule has a good stability.6. To study the effect of luodantiaozhi capsual on blood-lipid of hyperlipidemia rats. The hyperlipidemia model was made successfully after being fed by high fat feeder continuously for 15 days. Then they were lavaged for 30 days, gained blood by select the rats′eyeball.Determin blood setum TC, TG, LDL-C, HDL-C.The results reveal that luodantiaozhi capsual has the effect on adjusting blood fat.
