Background and Objective Biliary obstruction is a common disease in clinic,the causes of complications is choledocholithiasis ,bile duct injury and many other factors.it's may cause liver and other important organs damage.But when serum bilirubin start to increase? When will reache the peak?Whether does the jaundice aggravates with the time of biliary obstruction?We hope to carry out some research about the chage of serum bilirubin by the experiment of wister rats. To provide an evidence for clinical application.Method Choose healthy 72 wister rats, weight 180~220g, no limit for male and female, after one-week adaptive breeding,divide randomly them into biliary obstruction group(A1~A6) and the sham operation group(B1~B6), each group divide into 6 time points(1day,3 days,7 days,14 days,21 days,35 days), there are 6 rats in each time point.Model making:Rats preoperative fasting 12h, use 2% sodium phenobarbital 100mg/kg intraperitoneal anesthesia, clean operation, skin preparation by scissors and blade, surgical field disinfection byⅢAnerdian, upper abdomen incision, find the bile duct along the hepatoduodenal ligament, double ligation the bile duct above the duodenum,called biliary obstruction group, the sham operation group fractionated out the bile duct,no ligation, then abdomen-closing. the rats were killed after operation at 1day,3 days,7 days,14 days,21 days,35 days,each time point killed 6 rats. Take venous blood 3~4ml to detection the bilirubin and transaminase,take some liver to pathological examination .Data representation using x±s, mean comparison using one-way anova,group comparison using paired sample t test, p<0.05 have statistical significance.Result The TB of biliary obstruction group after operation are respectively 59.04±10.59(1D),72.23±24.07(3D),17.80±2.95(7D),15.95±31.33(14D),0.85±0.37(21D),0.61±0.07(35D);DB: 48.86±8.49,59.58±19.34,16.66±3.02,2.85±2.53,0.82±0.33,0.75±0.38; compared with the sham operation group ,after ligament bile duct 7days ,A group higher than B group (p<0.05),after 21 days A group lower than B group (p<0.05).IB: 10.21±2.11, 12.70±4.75, 5.14±7.40, 4.15±6.95, 0.24±0.16, 0.15±0.05, after operation 3 days A group higher than B group (p<0.05),after 21 days A group lower than B group (p<0.05);TBA: 145.07±20.63,92.20±8.32,61.38±5.39,30.55±36.00,17.1±8.27,14.03±3.26,after operation 3 days A group higher than B group (p<0.05) . ALT:1653.98±126.43, 529.78±56.42, 156.54±21.35, 136.34±21.35, 88.68±11.45, 50.39±4.45 after operation 14 days A group higher than B group (p<0.05) .Conclusion 1.The rats after ligament bile duct 1 day can be observed significantly increased, 3~7days reached the peak, then gradually decreased, after 21 days, the indicator significantly lower than normal.the reason still not clear at present.Using rats for reducing janudice experiment is inaccurate,because even without reducing janudice,the bilirubin also will decreased.after ligament bile duct, the indicator will significantly lower than normal, it's different from human, the mechanism need further research.Experimental Study on therapy of avascular necrosis of the femoral head with vascular endothelial growth factor2. Liver fibrosis of rats after ligament bile duct rats 7~14days reached the peak, 21 days decrease without for reducing janudice.
