Depression is mental disorder with high prevalence.The pathology mechanism is not be explained clearly nowdays.It is an global problems for mental healthy. Although anti-depressants have been used to treat depression,the effect is not so good. Acupuncture is a more effective therapy for depression,with less side effect.The mechanism for the efficiency of acupuncture on depression is not be explained very well.We hope to find some relative mechanisms on acupuncture anti-depression through studying neuronal cell apoptosis and neurogenesis and the results can be used for better explanation the mechanism of acupuncture anti-depression.ObjectiveTo explore the mechanism of electroacupuncture on chronic stress depression, we observed the behaviors for stress rats and the expression of Bcl-2 and GAP-43 in the brain of stress rats.MethodsForty Sprague-Dawley male rats were randomly divided into five groups, control group,control electroacupuncture,model group,electroacupuncture group and Pozac group.There were 8 rats in each group.(1) Chronic stress depression animal model was established and the behaviors were used to evaluate whether the model was successful.(2) Immunohistochemistry method was used to detect the expression Bcl-2 and GAP-43 in cortex and hippocampus.Results1.Chronic stress depression animal model was established,and Open field test, sucrose intake,and body weight balance were used to evaluate the model.After stress, compared with control group rats,the model group rats' crossing numbers,rearing times,the level of sacchar-consumption,body weight changes,were apparently less than those of control group rats'(P<0.05).In comparison with model group,the crossing numbers,rearing times,body weight changes,the level of sacchar-consumption,being apparently increasing in electroacupuncture group and Prozac group(P<0.05);those indexes were not apparently differences between electroacupuncture group and Prozac group(P>0.05).2.The expression of Bcl-2 in cortex data show that,compared with the control group,the expression of Bcl-2 in model group was more higher(P<0.05).The expression of Bcl-2 in electroacupuncture and Pozac group were higher than model group(P<0.05).The Bcl-2 expression was no different between electroacupuncture and Pozac group(P>0.05).The expression of Bcl-2 in hippocampus of model group was no difference from control group(P>0.05).Hippocampal Bcl-2 expression in electroacupuncture group was higher than model group(P<0.05).The Bcl-2 expression in hippocampus of electrocupuncture group was difference from Pozac group(P<0.05).3.Compared with model group,the expression of GAP-43 in cortex of the elcetroacupuncture and Pozac group were more higher,and there was significant difference between electroacupuncuture and model group(P<0.05).The expression of GAP-43 in cortex of Pozac group was almost the same level with the model group (P>0.05).Compared with the control group,GAP-43 expression in hippocampus of model group was lower(P<0.05).GAP-43 expression in hippocampus of electroacupuncture group,Pozac group was higher than model group,and the electroacupuncture GAP-43expression was significant different from model group (P<0.05).Conclusions1.The changes of behaviors on the animal model was successful,chronic stress could cause the rats to move less in open field test,suppress body weight gain of rats, decreasing in sucrose intake,but electroacupuncture can reverse it.2.Chronic stress could cause Bcl-2 expression decreasing,but electroacupuncture interfering could increase Bcl-2 expression in brain of stress rats, the results indicated that electroacupuncture could increase Bcl-2 expression may be had some relationship with behaviors changes of stress rats.3.Chronic stress could cause decreasing GAP-43 expression in brain of stress rats,whereas electroacupuncture could reverse it to protect the decreasing of neurogenesis under stress condition. |
