Fluorescence and bioluminescence imaging is a novel molecular imaging technique, which can be utilized to monitor in real-time and non-invasive the physiological, biochemical, and pathological processes in vivo. However, there are no systematic fluorescence-labeled animal models and cells available in China and resulted in that the technique was not used extensively in our country. The purpose of this study is to establish fluorescent and bioluminescent animals and cells that could be useful resource for bioresearch and medical research. In this study, the high-level expression GFP transgenic mice, LUC transgenic mice and 12 tumor cell lines were established successfully. We also test imaging mouse model using DsRed-labeled tumor cell line (B16-RFP) and GFP transgenic mouse. We utilized in vivo imaging system and fluorescence microscopy to monitor dynamically tumor in whole-body and cell level directly and we observed the inhibition effect of Lentinan on tumor with this technique, which verified the practicality and vivacity of the technique.
