Objective:By observing the effect of Ligustrazine on the patholog ical changes in kidney organization of Diabetic Nephropathy rats and the expression of TGF-β1,CTGF,investigate the mechanism of Ligustrazine in preventing diabetic glomerular sclerosis and interstitial fibrosis.Method:After creating the DN models of SD rats,divided the animals into four groups randomly:including model group,west medicine group, Ligustrazine(high and low-dosage)groups.Take another SD rats cut right kidney as normal group,total five groups.The rats of treating groups are affused in the corresponding medicine,the other are affused in the same volume water during 12 weeks.Test UAER in the 4th,8th,12th week respectively and measur the level of BUN,Scr,TC,TG in serum of every group 12 weeks later,then kill them,cut the left kidneys,fix,observe the pathological transformation of renal glomerulus.The CTGF and TGF-β1 are detected by immunohistochemistry and statistical analysis.Result:Comparing with the normal group,the level of TG,TC and UAER,BUN,Scr of the model group rose continuously(P<0.01;P<0.05). The expression of CTGF and TGF-β1 in renal organization raised significantly(P<0.01).Ligustrazine high dosage had the role of getting TG,TC and UAER,BUN,Scr decreased,and step down the expression of CTGF and TGF-β1 in renal interstitial(P<0.05).Conclusion:Ligustrazine could decrease the drainage of uric protein,prevent the renal interstitial fibrosis,postpone the process of DN.Turning down the TG and regulating the expression of TGF-β1,CTGF in kidney may possibly be one of its functional mechanisms.
