Chanru Granula(CG) is a traditional Chinese medicine containing six herbs:Radix et Rhizoma Ginseng,Radix Angelicae Sinensis,Radix Ophiopogonis,Semen Vaccariae and Radix Platycodonis.The medicine has functions of dispersing the depressed liver-energy to regulate the flow of Qi(vital energy),enriching the blood and Qi,and dredging the meridians to stimulate lactation.It is indicated for the treatment of periparturient hypogalactia syndrome,which is caused by deficiency of Qi and blood,or stagnation of Qi due to depression of the liver.The objective of this study investigates the pharmaceutics and pharmacology of CG.To investigate the optimal preparation processes of GC,Angelica oil was extracted by distillation,and the optimal process was screened with the factors experiment design.The alcohol extractive procedure of Ginseng was screened with orthogonal test,according to the content of Ginsenosides Rg1,Re and Rb1 by HPLC.The water extractive procedure of CG was selected by orthogonal test,according to the extractive rate,total saponins content and total polysaccharide content.The optimum molding conditions including excipient compatibility,wetting agent and desiccation temperature were selected by the factors experiment design.To establish the quality control methods for CG,Three berbs Ginseng, Angelica and Platycodonis were identified by thin-layer chromatography (TLC) method,and the content of Ginsenosides Rg1,Re,Rb1 of CG was measured by RP-HPLC gradient elution.The results revealed that the special spots of Ginseng,Angelica and Platycodonis were identified by TLC,The linear correlation of Ginsenoside Rglwas available between 0.272 and 4.080μg(r= 0.99997);Re was between 0.289 and 4.335μg(r=0.99994);Rb1 was between 0.342 and 5.130μg(r=0.99995).The average recovery of Ginsenoside Rg1 was 96.98% (RSD=1.74%);Re was 99.61%(RSD=2.76%);Rb1 was 97.38%(RSD=2.32%). The efficacy of CG in augmenting milk production was investigated in a rat model in which lactation was reduced by bromocriptine treatment. Postpartum rats were randomly assigned to normal control group,model control group,Ruquan Granula positive control group,low-dosage CG group, mid-dosage CG group and high-dosage CG group.The model control group was given with bromocriptine;TheRuquan Granula group with both bromocriptine and Ruquan Granula;The CG groups with bromocriptine and different dosages of CG daliy for 7 days after delivery.The lactational performance was assessed by weighing the pups.The concentrations of rat PRL in rat serum were measured by enzyme-linked immunosorbent assay(ELISA);The mammary and pituitary glands were examined at the 8th day after postpartum.The results revealed that the endogenous levels of rat PRL and the volume of milk production were significantly reduced by intragastric administration of bromocriptine everyday,lt is significantly different from normal control group(P<0.05).The serum prolaction(PRL) levels and the volume of milk production were significantly increased by intragastric administration of the high-dosage CG everyday.It is significantly different from model control group(P<0.05).These results expected that the preparation processes of CG were established,and the new methods of CG quality standards were simple and reliable.It increased milk production in a rat model.
