Objective: The purpose of this study was to observe the clinical experienced decoction (Sheng Ya Ning) which is composed on the basis of benefiting qi, astringing yin and stemming desertion, and cooling blood, removing stasis and resolving toxin, affected septic shock mice caused by LPS, and explore the mechanism of Sheng Ya Ning effect to septic shock by detecting TNF-α,iNOS and ET-1.Methods: SD mice were classified into 6 groups: blank control group, shock model group, Sheng Ya Ning group, Sheng Mai group, Xiao Ji group and positive control group. The SD mice were given LPS by tail vein injection to establish shock models and MAP (mean arterial pressure) was observed by BL-420E~+ biological function experimental system. After 2 hours, livers and lungs were collected to observe their pathological morphologic change and TNF-α,iNOS and ET-1 in mice livers and lungs were detected by immunohistochemical methods.Results: 1. The MAP of the shock model group was decreasing progressively after LPS injection; given treatment to all medicine groups, MAP decreasing of those groups was relieved, with Sheng Ya Ning group most prominently. 2. Shock model mice had obvious inflammatory pathological changes in livers and lungs, and even stasis; given treatment, pathological damage of all medicine groups was relived in different degrees and Sheng Ya Ning groups relieved most prominently. 3. Compared with blank control groups, TNF-αexpression in livers and lungs of shock model group increased significantly (P<0.01); Given treatment, TNF-αexpression in livers and lungs of all treatment groups decreased remarkably (P<0.01) and the expression of Sheng Ya Ning group was the lest. 4. Compared with blank control groups, iNOS expression in livers and lungs of shock model group increased significantly (P<0.01); given treatment, iNOS expression in livers and lungs of all treatment groups decreased remarkably (P<0.01) and the expression of Sheng Ya Ning group was the lest. 5. Compared with blank control groups, ET-1 expression in livers and lungs of shock model group increased significantly (P<0.01); Given treatment, ET-1 expression in livers and lungs of all treatment groups decreased remarkably (P<0.01) and the expression of Sheng Ya Ning group was the lest.Conclusions: 1. Sheng Ya Ning can mitigation the progressive blood pressure decrease by LPS of septic shock mice; 2. Sheng Ya Ning can relieve the pathological damage in livers and lungs of septic shock mice; 3. Sheng Ya Ning can inhibit the expression of inflammatory mediator TNF-αin livers and lungs; 4. Sheng Ya Ning can inhibit the expression of iNOS in livers and lungs; 5. Sheng Ya Ning can inhibit the expression of endothelin-1 in lungs and livers.
