Rhubarb(Dahuang in Chinese) is widely used in clinic.Although Rhubarb is not toxic traditionarily,some adverse reactions on rhubarb are reported in recent years.Rhubarb tips in some area may have some toxic effects,at the same time may be related to clinical drug-related misconduct.Jun Da huang for the capture of strong materials,such as the spleen and stomach weak cold should be used with caution.The risk of adverse reactions may increase if the constitution of patients does not classified.For example,children and the elderly physical relatively are weak,adverse reactions may occ ur if the dose of Rhubarb to them according to the normal adult dosage,.In this paper,the national "Tenth Five-Year" key scientific and technological projects - " the demonstration of adverse reactions on Rhubarb and their safety evaluation of compound preparation "(NO.2004BA721A14) under the framework,at the higher dose of the full exposure of the toxic side effects of Rhubarb through comparative study of normal minors Rhubarb and the safety of aged rats and differences in toxicity study Rhubarb correlation with age,and for different age groups to provide the clinical basis for rational drug use.1.The effects in rats of rhubarb tolerance3-week-old SD rats fed by garage a minor to give the total extract of Rhubarb were observed after administration of 1 week,2 weeks,5 weeks after drug withdrawal. after a month,the results show that:(1) the dose of Health Rhubarb mice did not lead to the death of a minor;(2) the group of low-dose generally normal,body weight increase faster than the blank group,individual biochemical abnormalities,liver index increased,abnormal liver pathology occurred(3) in the dose group of animals generally poor weight than the control group no significant increase in the trend of individual biochemical abnormalities,liver index increased,abnormal liver pathology did not occur.(4) the group of high dose of are generally poor,slow weight gain to adult body weight lower than the normal rats administered individual biochemical abnormalities,index of liver and kidney increased,(5) after the withdrawal of low-dose group,middle dose group of animals body weight tended to increase more than the blank group,high-dose group of animals weighing less than the blank control group,but no significant difference is found between them.Every group of animals are generally normal with urine and mental activity back to normal.2.The effects in the age of rats Rhubarb tolerance1 year or older SD rats fed by gavage to give the total extract of Rhubarb were observed after administration of 1 week,2 weeks,5 weeks after drug withdrawal after a month of a comprehensive animal,the results show that:(1) blank control group and low dose group had no death,died in the occasional dose group,high-dose group were killed,and the older are more easily killed.(2) low-dose group generally normal, with normal body weight,individual biochemical abnormalities,no abnormal liver pathology.(3) the middle-dose group of animals generally is worse,with normal body weight,individual biochemical abnormalities,no abnormal liver pathology.(4) high dose group of animals generally worst,weight loss,prolonged administration of albumin with decreased delivery of early transaminase,creatinine,blood urea nitrogen transient increase in IL-5 weekend -6(with the blank group comparison,p<0.05),IL -8(compared with blank control group,p<0.01) were increased,abnormal liver pathology was not observed.3 Aged rats on the toxicity of Rhubarb toleranceExperimental results show that side effects of Rhubarb on rats are age-related.In the same dose,the minors were not obvious,rat pathological changes in organs,animals in general,the blood biochemical indices were normal,and so on;and aged rats can be seen varying degrees of toxicity,and the occurrence of side effects were dose-related and can be seen in the dose group of animals generally deteriorated,individual biochemical abnormalities,high-dose group of animals generally poor,a significant drop in body weight,the majority of biochemical abnormalities,liver biopsy can be seen to change.In addition,the experiment found that the order of aged rats killed was a negative correlation with body weight,that is,the greater weight of the animals died sooner.Acquired as a result of this experiment would thus be that the older the animals are,the toleration of Rhubarb may be worse.4.the proposed use of Rhubarb between different individual age groupsT he body of minors have the developmental aspect of the imperfect,but at the same time they are vitality and Rhubarb appropriate dose may not increase its damage.Thus when clinical application of Rhubarb to children,without causing severe diarrhea, an appropriate increase in Rhubarb dose perhaps will have a better effect.Clinical application of rhubarb to aged subjects might be considered to reduce the dose appropriately,and great attention should be paid to a variety of possible toxic or adverse effect.For long-term application,it could be used from a lower dose firstly, then raise the dose to normal level gradually when the subject has the tolerance.
