Phlegm Stasis, Liver And Kidney Tonic Side Comparison Of High-fat Rat Model Of Lipoprotein Receptor Expression

Objective:To study the effect of QuTanHuaYu Fang and BuYiGanShen Fang on blood-lipid of animal model of hyperlipidemia in rats and to study the different expression of them on high-density lipoprote in recepter low-density lipoprotein recepter.Clarify different herbs can work on Hyperlipidimia through multiple targets and channels which can interfere with the process of cardio cerebrova- scular disorders and artherosclerosis.Methods:There are 60 Wistar male rats, rats were randomly divided into five groups, the normal group, the model group, QuTanHuaYu Fang group ,BuYiGanShen Fang group and medicine group. Rats in the normal group were fed with general feed stuff, while rats in the rest group were all given feedstuff with rich fat and established animal model of hyperlipidemia in rats.4 weeks after modeling,the treatment group were given corresponding gavage of the drug , model group and control group were given equivalent gavage of the normal saline. For 30 days after administration,blood serum TC,TG,LDL-C,HDL-Cdetermination in rats on full-automatic biochemistry analyzer with enzymatic reaction.The levels of high- density lipoprotein recepter of the livers of the rats were detected by RT-PCR. The levels of high-density lipoprotein recepter and low-density lipoprotein recepter of the livers were detected by Western-Bloting.Results:QuTanHuaYu Fang and BuYiGanShen Fang have obviously reduced the concents of blood TC, TG,All the traditional Chinese medicine groups had a less effect and notable difference to medicine group (P>0.05). QuTanHuaYu Fang and BuYiGanShen Fang have obviously reduced the concents of blood LDL-C.The difference were not remarkable and the effect of QuTanHuaYu Fang was best. QuTanHuaYu Fang and the model group have significantly increased the concents of blood HDL-C,and there was significant difference to the model group (P<0.01). There was not significant difference between BuYiGanShen Fang and the normal group (P>0.05). BuYiGanShenFang increase HDL-C significantly effect. Each treatment group that high-density lipoprotein receptor SR-BI gene expression and protein expression levels of rats were significantly lower than the model group, especially BuYiGanShen Fang optimal effect.Each treatment group of rats which LDL receptor protein expression levels were significantly increased to the model group, and QuTanHuaYu Fang group with western medicine group was no obvious difference(P >0.05).Cunclusions:QuTanHuaYu Fang and BuYiGanShen Fang all can regulate the blood-lipid, and to effectively improve the liver receptor HDL-C and LDL-C receptor expression, it reveal that QuTanHuaYu Fang and BuYiGanShen Fang can regulate blood-lipid and prevent atherosclerosis role of the different target .
