Objective:To investigate the protective effect of the retinal M(u|¨)ller cells in vitro in hypoxia and TGF-β2 intervention,furthermroe,its protection mechanism of action was studied at the same time.Methods:1.Modified enzyme-digesting method for culturing retinal M(u|¨)ller cells of Newborn-rat;2.Make anoxic experimental model through joining with sodittm dithionite(final concentration of 1.0 mmol/L) in the medium;3.To observed ultramicrostructure of RMCs of Newborn-rats in the serum of BSHX decoction in different conditions;4.Glutamate up-take activity of M(u|¨)ller cells was determined,using isotopic techniques,by intracellular[3H]labeled D, L-glutamate concentration;5.To detected the activity of glutamine synthetase(GS) and the Lactate dehydrogenase(LDH) quantity released into the incubation medium from cultured M(u|¨)ller cells with the Spectrophotometer.Results:1.Retinal M(u|¨)ller cell ultrastructure had obvious pathological damages in hypoxic conditions,the serum of BSHX decoction can conspicuous reduce pathological damage;2.the serum of BSHX decoction can significantly enhance the GS activity and the Lactate dehydrogenase(LDH) quantity of retinal M(u|¨)ller cells in hypoxia and(or) in the intervention of TGF-β2;3.In hypoxic conditions retinal M(u|¨)ller cells glutamate intake increased,the serum of BSHX decoction can increase glutamate uptake activity of M(u|¨)ller cell,but in hypoxia and(or) in the intervention of TGF-β2,glutamate intake has decreased.Conclusion:1.BSHX can reduce the M(u|¨)ller cells' injury in hypoxic conditions 2. The effects of BSHX on LDH and GS maybe a protection mechanism for DR;BSHX may be useful for the therapy of DR;3.BSHX can enhance RMCs glutamate uptake activity and lessen glutamate excitotoxicity,when hypoxia and TGF-β2 conditions exist at the same time,the serum of BSHX decreased RMCs glutamate uptake capacity may be a self-protective effect in a single cell environment.
