Objective : To study the Glutamate up-take activity of M端ller c ell and the LDHquantity released from M端ller cell in normal and high glucose; to investigate the effect of The serum with BSHX decoction to the M端ller cell in normal and high glucoseMethods: 1. Modified enzyme-digesting method for culturing M端ller cell of Newborn-rat 2.serum with BSHX decoction was gained by the serum pharmacology method of traditional Chinese medicine .3 To detected the optical density of cultured M端ller cell of Newborn-rat in the serum of BSHX decoction at different concentration in high glucose with MTT, to determine the effective concentration of the BSHX decoction - serum 4. To detected the the Lactate dehydrogenase (LDH) quantity released intothe incubation medium from cultured M端ller cell with the Spectrophotometer.5. Glutamate up-take activity of M端ller cell was determined , using isotopic techniques , by intracellular [~3H]labeled D, L-glutamate concentration.Results: 1 .Glutamate up-take activity of M端ller cell increases can be causedrapid ly by high glucose , however this effect is temporary , a sharp decreases hapen to it with the time passing 2. the BSHX decoction - serum was discovered to be able to enhance the Glutamate up-take activity of M端ller cell in normal glucose , meanwhile this beneficial effect afforded by the BSHX decoction - serum were found to be the strongest after 24 hours. 3 . the BSHX decoction - serum was discovered to be able to enhance the Glutamate up-take activity of M端ller cell in high glucose , meanwhile this beneficial effect afforded by the BSHX decoction - serum were found to be the strongest after 48 hoursConclusion: 1. enhancement of glycolysis in M端ller produced by high glucose In shorttime may be one of the mechanism of increases of Glutamate up-take activity of M端Her cell in high glucose 2. retinal M端ller cells in culture maybe are resistant to high glucose, however persistent high glucose can result in injury of retinal M端ller cells.3. the BSHX decoction - serum was discovered to be able to inhibition the down regulation of Glutamate up-take activity of M端ller cell ,whitch maybe one of the mechanisms that BSHX method can prevent and cure DRP...
