Periplaneta Americana have wide-ranging pharmacological effects and unique pharmacodynamic actions . Both documents and modern pharmacological research indicate that Periplaneta Americana have effects on ulcerative colitis(UC) , hepatitis and cirrhosis. Its application is also developing. Active fraction of anti-UC and anti-hepatic injury was screened by in vivo experiments, then the extraction and purification process was studied, the efficacy of purification product was verified and the standardization of active fraction was also studied in this project, which present as following.First chemical composition identification of Periplaneta Americana was conducted , then in vivo experiments of mice was used to screen the anti-UC and anti-hepatic injury active fraction .The result revealed that 70% and 90% ethanol extracts were active for anti-UC , whose amino acids and peptides were analyzed and separated , and the water extract was active for anti- hepatic injury,whose main ingredients were proteins and peptides.The extraction process of anti- hepatic injury was studied and optimized:the result of comparing three extraction methods was that alkaline water extraction was best; main factors were systematically investigated,the extraction effect was good when solid-liquid ratio was 1:25, extraction time was 60 min, extraction pH value was 9 , extraction temperature was 40℃.The purification process of anti-hepatic injury was studied and optimized: compared with alcohol sedimentation technique and isoelectric precipitation technique,foam separation technique had advantages .A single-factor experiment was conducted to inspect the process parameters and optimal conditions were achieved finally. The foam performance was good when gas flow rate was 600mL·min-1, initial feed concentration was 0.45 mg·ml-1 , initial pH value was 4.5 ,feed volume was 1000ml and room temperature was kept. The recovery was about 67% and total protein purity was 58% in the production(29.95% in raw materials).The efficacy of foam product was verified by in vivo experiment and the standardization of active fraction was studied ,which included macroscopic identification,physical and chemical identification, TLC identification and content determination.
