OBJECTIVESelect the appropriate concentration of leech extract which can highly inhibit the proliferation of RF/6A cells in vitro.METHODS1.rhesus choroid-retinal vascular endothelial cells(RF/6A) were cultured in vitro,and the cultured cells were identified by morphological examination and theⅧfactor fos-related antigen,immunohisto-chemistry techniques.2.RF/6A cells were exposed to leech extract with different concentrations (128,64,32,16,8 and 0mg/ml) for 24 hours,Colorimetry MTT was used to detect the viability of RF/6A cells in different concentrations,got the OD values and calculated the 50%inhibitory concentration(IC50) of leech extract.3.Selected the appropriate concentration that inhibits RF/6A Cells proliferation greatly and without any obvious toxic effect.RESULTSAdministration of 32~128mg/ml leech extract for 24hours could significantly inhibit RF/6A proliferation(P<0.05).However,the 128mg/ml concentration had an obvious toxic effect which detected by morphological examination.The 64mg/ml concentration's inhibitory proliferation is superior.CONCLUSIONCultured RF/6A cells appear to have certain proliferative abilities.There is a remarkable inhibitory effect of leech extract on RF/6A cells.This experiment provides an experimental evidence to counter and prevent the the growth of new vessels with medicinal herbs.
