Objective:under the environment of high glucose condition to explore whether the Albizia julibrissin extract(AJE)have an effect on H?man retinal vascular endothelial cells(HRCECs)proliferation and it's expression of VEGF,on this basis further clarify whether the Albizia julibrissin extract can be used in the treatment of diabetic retinopathy.Methods:The h?man retinal vascular endothelial cells were cultured in a high-glucose environment and identified by cellular immunofluorescence technique.After determining the experimental concen tration of Albizia julibrissin extract thro?gh preliminary experiment,MTT colorimetry assay was used to study the effect of different concentrations of Albizia julibrissin extract on the HRCECs proliferation.Protein western blot was used to definite the effect of the different concentrations of acacia bark extract on the expression of VEGF in the HRCECs.Results:1.In the high-glucose environment,due to the intervention of different concentrations of Albizia julibrissin extract,the level of cell proliferation will be inhibited to varying degrees.1)If the cells cultured in the same time,the n?mber of HRCECs in the lows?gar group was significantly reduced compared with that in the group of high-glucose,P<0.05,which was statistically different.2)If the cells were cultured by 24 hours or 48 hours,the inhibition rate of HRCECs is increase accompanied by the rising levels of con centrations of Albizia julibrissin extract(10?g/ml,20?g/ml,30?g/ml,40?g/ml)+ high-glucose group,which compared among different groups,the results showed significant difference(P<0.05).But thro?gh 72 hours,absorbance of different concentration of Albizia julibrissin extract(10 ?g/ml,20 ?g/ml)+ high-glucose groups have no obvious differences in statistics,either in groups of different concentration of Albizia julibrissin extract(30 ?g/ml,40 ?g/ml)+ high-glucose,(P > 0.05),no statistical significance,therefore in the next experiment we do research on 24 hours and 48 hours;2.Western Blot was used to detect the expression of VEGF in HRCECs of different groups in this experiment: 1)After 24 hours or 48 hours culture,the expression of VEGF in HRCECs of the high-glucose group was higher than that of the low-s?gar group,and the results of data analysis were significantly different,which has statistical significance(P<0.05).2)Cultured in the the same time,the expression levels of VEGF is decrease accompanied by the decrease of Albizia julibrissin extract concentrations,which compared among different groups(Albizia julibrissin extract(10?g/ml,20?g/ml,30?g/ml,40?g/ml)+ high-glucose),and the results were significantly different(P<0.05).Conclusion: High glucose environment can promote the proli feration of HRCECs and increase the expression of VEGF in vitro.The proliferation of HRCECs cultured in vitro in a high glucose en vironment was inhibited by the extracts of acacia bark at different concentrations.At the same time,the expression of VEGF in HRC ECs can be down-regulated by the intervention of different concentrations of Albizia julibrissin extract.
