Abstract/Summary:
Objective To reveal the influence of electro-acupuncture(EA) therapy on the synaptic plasticity of hippocampal neurons of senescence-accelerated mouse(SAMP8) and partly explore the mechanism of EA on Alzheimer's disease(AD) from neural cell adhesion.Methods SAMP8 was taken as animal model of AD.The treatment of acupunctur on Baihui(Du20) and Yongquan(Kid1) with EA was applied once daily and lasted for 21days.After treatment,the changes of learning and memory abilities of SAMP8 were observed and recorded by Morris water maze and used to evaluate the curative effect of EA therapy.The expressions of Neural Cell Adhesion Molecule(NCAM) and Nuclear Factor kappaB(NF-kB) in hippocampal neurons were detected by immunohistochemistry and Instiu hybridization.Results 1.Comparing with control group,the average escape latency of EA group were obviously decreased(P<0.05).Comparing with control group(30.59±6.02s),the times of swim in platform quadrant of EA group(38.55±3.59s) were obviously increased (P<0.05).2.Comparing with control group,the expression of NCAM and NF-kB of EA group are significantly increased(P<0.05).Conclusion 1.EA could improve the learning and memory abilities of SAMP8 in a certain extent.2.EA could increase the expression of NCAMa nd NF-kB in hippocampal neurons effectively.3.Its cellular molecular mechanism may be related to promote adhesion and synapse plasticity of neurons by inducing the composition and expression of NCAM and NF-kB.
