Xinqingning Capsule is a new compound preparation of traditional Chinese Medicine,which is composed of Radix Salviae Miltiorrhiae,Radix Notoginseng, Flos Carthami,Broneolum Syntheticum,Comu Bubali and Calculus Bovis. Boasting of the function of activating blood circulation to dispel the blood stasis and opening the orifices to alleviate pain,it can be used to treat coronary disease, angina pectoris,chest distress,palpation and vertigo.Under the guidance of the theory of the traditional Chinese Medicine,modern techniques have been utilized to research the preclinical study of this capsule.On the basis of the clinical requirement,combined with the nature of the drugs in the prescription,capsule was chosen to be the form.According to the action of the prescription and the physico-chemical properties of the ingredients in the drugs, orthogonal design have been applied to select the optimum conditions,which were as follows:Broneolum Syntheticum,Comu Bubali and Calculus Bovis were pulverized and sieved with 100-mesh sieve,reserved.Radix Salviae Miltiorrhiae and Radix Notoginseng were extracted by 70%ethanol for three times:8 times of ethanol for 3 hours for the first time;6 times of ethanol for 2 hours for the second time;4 times of ethanol for 1 hour for the last time.The extract was merged and infiltrated by 400-mesh filter cloth,then the ethanol was recycled and the leftover was concentrated until the density of which reached 1.10(60℃).Radix Salviae Miltiorrhiae and Radix Notoginseng,which were extracted with ethanol,was then extracted with 10 times of water twice,together with Flos Carthami.The water-extract was incorporated and high-speed centrifugated(8000r/min).The supematant was concentrated until the density of which reached 1.10(60℃).The two extracta were put together,and spray drying and one-step granulation techniques were utilized to dry and shape.Then the quality standard of the preparation was studies.Salvianolic acid B in the principal drug Radix Salviae Miltiorrhiae was determined by HPLC method.Meanwhile,TLC method was used to identify Radix Salviae Miltiorrhiae,Radix Notoginseng,Flos Carthami,Broneolum Syntheticum,Comu Bubali and Calculus Bovis.On the foundation of the controllability of the preparation,the rudiment stability of it was also studied.And the results showed that the technique was reasonable and feasible;the quality standard was controllable;the quality of the end item was generally stable.
