Abstract/Summary:
Objective:To research on core clock genes' negative feedback mechanism of acupuncture on rhythm phase,and provided evidences in molecular biology on the regularity of acupuncture's effect on different hours.Methods:Applied the methods of Aschoff' entrainment and the technology of quantitative RT-PCR,we study on the spontaneous behavior circadian rhythm of mice by electro-acupuncturing "Baihui" (Du20),changqiang"(Du1),and research mice per1,cry1 and cry2 mRNA levels in SCN occurred at CT6.Results:electro-acupuncture circadian-depended regulated the acrophase of mice spontaneous behavior circadian rhythm.In constant darkness electro-acupuncture induced phase advance at CT6,it also elicted phase delay at CT18 in mice spontaneous behavior circadian rhythm.Inhibitory effect of electro-acupuncture on mice per1 and cry1 mRNA levels in SCN occurred at CT6,and it was a downtrend on the genetic expression of cry2 mRNA at the same time.Conclusion:Electro-acupuncture is one kind of nonphotic zeitgeber.It down regulated the genetic expression of per1,cry1 and cry2 mRNA at the noon of subjective daytime,thus,reduce it's negative feedback inhibition on positive molecule,to achieve activating arousal mechanism,improving activity,advancing spontaneous motor circadian acrophase.
