Lapemis hardwickii and Hydrophis cyanocinctus,as the major breed of seasnake in our country,are habitually used to treat rheumatics,quadriplegia, arthralgia,acariasis,infirmity,and malnutrition of the children.At the present time,most of the related studies of seasnake are mainly focused on the snake venom,however,the information on the other chemical constituents of seasnake is still scarce.In due to find the constituents that relate to pharmacodynamic action,we studied the chemical of Lapemis hardwickii and Hydrophis cyanocinctus with the pharmacodynamics.seasnake were extracted two times with 50%ethanol,then the concentrated extract(SS-70E) was suspended in water and partitioned successively with petroleum ether(SS-P),ethyl acetate(SS-A) and n-butanol(SS-B).Each part was concentrated under reduced press.The activity of every layer including the residues of aqueous solution(SS-W) was evaluated by the pharmacological tests. The clearance of carbon particles and the weight of immune organs of mice were determined.The fact was shown that SS-A could put on the weights of mice's immune organs,improve the phagocytic function of reticuloendothelial cell and enhance the immunity function of mice.Further invastigation on the chemical constituents of the extract of SS-A by chromatographic methods,6 monomers have been isolated and 5 monomers were identified by physico-chemical properties and spectral methods.They are cholesterol(SS-1),1-methylhydantoin(SS-2),p-hydroxyphenyi acetic acid(SS-4), methyl p-hydroxybenzeneacetate(SS-5) and a new ceramide(SS-3).Among them, compound SS-2 is for the first time isolated from halobios,compound SS-4,SS-5 are for the first time isolated from seasnake.Morden research has shown that 1-methylhydantoin possesses extensive actions,such as antiinflammatory,relieving cough,preventing analbuminemia, eliminating active oxygen and free radicals.The effects on immune system are in line with the pharmacological tests.So a sensitive and specific NP-HPLC method has been developed,for the first time,to quantify the bioactive chemical component.Meanwhile because seasnake has a great amount of cholesterol,we also adopt TLCS method to quantify the compound.The results showed that the quantitative method is reliable,precise,simple and accurate.What we have done are helpful for rational development and utilization of seasnakes and can offer effective methods for quality control of seasnakes.At last,TLC and SDS-PAGE test have been carried out to differentiate seasnakes and other marine animals,seasnakes and terricolous snakes.The investigation are prepared for subsequent research.
