An Investigation on Chemical Constituents and Quality Control of Stellaria dichotoma L.var.lanceolata BgeRadix Stellaria dichotoma L.var.lanceolata Bge is a traditional Chinese crude drug, known as "yin-chai-hu" and recorded in the pharmacopoeia of P.R.China (2000 edition). It is mainly distributed in Ningxia Province. It tastes sweet and is cold in nature. It is considered capable of eradicating asthenic fever, and is used to treat fever due to deficiency of yin, comsumptive fever and fever in infantile malnutrition.During the past decade, many studies on Stellaria dichotoma L.var.lanceolata Bge had got great achievements. This chemical investigation was carried out for a better method to evaluate the quality of the crude drug and quality control.Chemical constituents of Stellaria dichotoma L.var.lanceolata Bge were systematically investigated, eleven compounds were isolated from the alcohol extract by using silica gel, macroporous resin, Sephadex LH-20,ODS chromatography and pre-HPLC. By employing physical and chemical properties ,modern spectrometric analysis techniques such as UV,IR,EI/MS,FAB/MS,NMR,13C-NMR,DEPT,1H-1HCOSY,13C-1HCOSY, seven compounds' structures of them have been elucidated. They're stigmast-7-en- (3β,5α,24s)- 3-Oglu],stigmast-7-en-3-ol,(3β,5α,24s)-, stigmast-5-en-3-ol,(3β)-, stigmast-7-en-3-ol,(3β,5α)-, Stellarine C, Stellarine B, Stellarine D. Two possible new compounds named as Stellarine C and Stellarine D belong to β-carboline alkaloids, and are isolated from the specie for the first time.Stigmast-7-en-3-ol,(3β,5α,24s)- has been chosen as the reference substance for qualitative and quantitative analysis of the drug for its function of "heat-clearing " and content. Therefore, a new TLC method and new HPLC-ELSD method were established. The result from 10 samples by HPLC-ELSD method determine that the minimum content of Stigmast-7-en-3-ol,(3β,5α,24s)- should be 0.020%. The HPLC-ELSD method is available, possible and accurate. Including those above, a protocol of the quality standard of the crude drug, Stellaria dichotoma L.var.lanceolata Bge, has been elaborated.As a supplement to the standard, Stellarine C, a specific chemical constituent and another reference substance, has been analyzed by HPLC for quantitative determination in 10 samples. The content of Stellarine C is about 0.009-0.073 mg/g. We have made a preliminary experiment on the HPLC fingerprints, and find out twelve common peaks in 10 samples. The result can be a base for the further qualitative or quantitative analytical research.
