Objective1. To investigate the inhibition of Pyruvate on cataract .2. To investigate the effect of Pyruvate on ultrastructure of lens in diabetic cataract.3. To investigate the effect of Pyruvate on GSH and MDA of lens in diabetic cataract .4. To investigate the effect of Pyruvate on ATP of lens in diabetic cataract .Methods1. Diabetic cataract was induced by glucose and containing 10% fetal calf serum in the culture medium MEM .2. The opacification of lens was detected by microscope , and the ultrastructure of lens was detected by transmission electron microscope .3. At the end of experiment, the level of GSH (glutathione), MDA (malondialdehyde), ATP in the lens were determined .4. Statistical analysis SPSS10.0 was used , with a P < 0.05 considered statistically significant .Results1. Pyruvate have protective effect to the caratact induced by glucose , to prevent high concentrations of glucose on the rat lens damage .2. Under transmission electron microscope , no changes were presented in control group , but mild changes were showed in treatment group and remarkable changes were found in model group .3. The contents of MDA were increased and GSH were decreased in model group compared with control group and the application of Pyruvate decreased the levels of lipid peroxidation ( MDA ) and increased the levels of GSH in comparison with model group . Conclusion1. Pyruvate can significantly delay or inhibit the formation of diabetic cataract.2. Pyruvate can significantly reduce oxidative damage of lens , and improve the lens of energy metabolism .
