Objective: To investagate the effects of Cornus officinalis polysaccharides on the anti-brain aging and the expression of nerve growth factor mRNA in aging model rats induced by D-galactose.Methods: The forty-three Wistar rats were randomly divided into 4 groups , the youth compares group(12), the senile model group(11) and two model medication administration groups(20). Senile model group and two model medication administration groups were daily injected D-galactose for 42 days in lateral region of abdomen to induce aging model . And two model medication administration groups were respectively perfused with 0.14g/kg,0.28g/kg Cornus officinalis polysaccharides for 30 days after making aging models. Measured the capability of learning and memory of rats , the content of MDA,LF, the activity of SOD,MAO-B,AchE in brain tissue , and the expression of NGFmRNA, as soon as observed the effect of Cornus officinalis polysaccharides on the above mentioned index.Results: Compared with the young group , the senile model group the content of MDA,LF, the activity of MAO-B,AchE were increased (p<0.01). But the capability of learning and memory , the activity of SOD , the expression of NGFmRNA were decreased significantly (p<0.01) . Compared with the senile model group , the capability of learning and memory , the activity of SOD , the expression of NGFmRNA were increased significantly (p <0.05, p<0.01) in two model medication administration groups , while the content of MDA,LF, the activity of MAO-B, AchE were decreased (p<0.05, p<0.01). The effects of the polysaccharide large dose group is more obvious than small dose group .Conclusion: Cornus officinalis polysaccharides could enhance the capability of learning and memory and enhance antioxidation, inhibit lipid peroxidation,raise the activity of aging-correlated enzyme and the expression of NGFmRNA,so it plays certain role in anti-brain-aging . The effects of the polysaccharide large dose group is more obvious than small dose group .
