Acupuncture Combined With The Rat Insulin-like Growth Factor In The Experimental Climacteric Syndrome

Purpose:PMS is an integrated symptom of endocrine imbalance and vegetative nervous disorder which is caused by perimenopausal ovary function progressive decay,the reduction of estrogen secretion and the reduction of progestational hormonesecretion or even stop.PMS has a very high incidence and various complications,which had become a severe disease of impacting women healthy and life quality and lifes,therefore,seeking a kind of safe and high efficient treatment has become a hot point of medical researcher.The aim of this study was to determine the effects of electroacupuncture associated with estrogen ovariectomized rats.This investigation studied the influence of electroacupuncture for serum E₂,FSH,LH,liver IGF-Iand liver IGF-Ⅱovariectomized rats so as to provide the theoretical basis for electroacupuncture in the clinical treatment of PMS.Methods:Divide the 60 Wistar female rats into 6 groups randomly:general group,sham group,ovariectomized control group,estrogen group,electricacupuncture group, electricacupuncture and estrogen group.Take ten of the Wistar female rats as general group without any treatments and take sham operation that only cutting off the fat organization which is around the ovary and then suture and disinfect on another ten.The others 40 rats cut the both sides of ovarys for making models.After the operation,injecting the penicilin (4U/ml)into the celiac each day,totally last 3 days,for carrying out the anti-inject treatment per day.according the normal feed,keep up vagina slice test under microscopefor 5 days after one week.The cells that vagina take off havn't appear the periodicity change, namely emasculationed,nomal feeding for 3 months and it came into being surrounding menopause model.estrogen group:treating after 3 months,ues nilestriol,put it into dust, dissolve it into the brine,feed by stomach acus,according to the inverse proportion of human and rat,provide the nilestriol 0.04mg/kg,isotonic Na chloride 10ml/kg each day.dissolve it, one time per day,10days as a period of treatment,during the periods of treatment stop one day,totally for 3 periods.Electroacupuncture group:beginning to electroacupuncture after 3 months,chooseShenshu,Taichong,Zusanli,Sanyinjiao(ambi),ues G6805Ⅱ electroacupuncture instrument,take on the continuum wave shelves,15HZ,one time per day, 15minites per time,10days as a period of treatment,during the periods of treatment stop one day,totally for 3 periods.the electroacupuncture treat as same as electroacupuncture group. general group,sham group,ovariectomized control group,without any treatment.All the animals feeding in the same condition.After 3 periods of treatment all the animals fast at the last treatment.24hours later,phlebotomize from the abdomen artery.use RIA.The rats were sacrificed,then taken out the liver.Used immunohistochemistry method to investigate the expression of IGF-Ⅰand IGF-Ⅱ.Results:1.Serum e2 levels:compared with the general group and sham operation group, the experimental peri-menopausal syndrome rats capacity of serum E₂ have obvious decreasing(P＜0.05)2.FSH and LH levels:compared with the general group and sham operation group,the experimental peri-menopausal syndrome rats capacity of serum FSH and LH have obvious increasing(P＜0.05).3.Compared with the general group and sham operation group,the experimental peri-menopausal syndrome rats capacity of liver IGF-I have obvious decreasing(P＜0.05).The capacity of liver IGF-Ⅱhave obvious increasing.4.Serum E2 levels:Electroacupuncture,estrogen,and Electroacupuncture and estrogen treatment can increase the experimental peri-menopausal syndrome rats serum E2 capacity and decrease the serum FSH and LH.compared with the robust model,the estrogen Group,Electroacupuncture Group has a significant differences(P＜0.05);estrogen Group, Electroacupuncture Group compare with the Group of Electroacupuncture and estrogen comparison,there were significant differences(P＜0.05)group of estrogen and Electroacupuncture compare with estrogen group,has no significant differences(P＞0.05);5.Electroacupuncture,estrogen,and Electroacupuncture and estrogen treatment can adjust the capacity of liver IGF-Ⅰand IGF-Ⅱ.Compared with the model group,the former 3 groups have significant different.(P＜0.05),and the capacity of liver IGF-I has significant increasing,IGF-Ⅱhas significant decreasing,which is in the Group of Electroacupuncture and estrogen and the estrogen Group.There are no significant different between the Electroacupuncture group and estrogen Group.Conclusion:Estrogen and electroacupuncture joint applications can significantly increase serum E2 levels,FSH and LH levels are significantly lower than single estrogen treatment achieved good results;Separate injections can improve the use of Electroacupuncture in serum E₂ and liver IGF-Ⅰ,IGF-Ⅱlevel for FSH and LH levels lowered But with the increase in Electroacupuncture and estrogen treatment group or estrogen treatment group compared role results clearly low.Electroacupuncture to increase emasculation mouse of dependence serum E₂ and liver organizations IGF-Ⅰ,IGF-Ⅱexpression of a certain Peri-menopause syndrome improve.Only effective treatment for menopause and the role Peripossible upward E₂,ICF-1 expression mechanism achieved.
